In a test applied 0.1 mL of undiluted this substance to eyes of 3 rabbits (OECD TG 405; GLP), average scores in 3 animals at time points of 24, 48, and 72 hours after application were 1 for corneal opacity, 0.4 for iritis, 2.0 for conjunctival redness, and 0.9 for chemosis. And it was assessed as highly irritating from a score of 1 in corneal opacity and scores of 2 and 3 in conjunctival redness observed in 2 out of 3 animals after 23 days. (SIDS (2004))
Threfore, it was classified in Category 2A.
Besides, another test applied undiluted this substance to eyes of 3 rabbits reported that produced slight conjunctival irritation, and slight and transient corneal injury reversed within a few days to 1 week. (ECETOC 95 (2005))

- - In a micronucleus test using bone marrow after an oral administration to mice (in vivo somatic cell mutagenicity test), the slightly increased frequency of micronuclei in animals in a high dose group (2000 mg/kg) was statistically significant in the first test but was not significant in the second (indicated a similar tendency).
The author of this study attributed this fact to hypothermia. Causing hypothermia only in animals at the high dose and increased micronuclei as its indirect effects were clarified by other chemicals.
It seems reasonable to conclude that the negative in vitro results and the equivocal in vivo results at a very high dose level that may be due to physiological stress indicate that propylene glycol phenyl ether does not pose a genotoxicity hazard at a possible exposure dose in the environment (SIDS (2004)).
From the above information, it was classified as "Not classified."
Besides, an Ames test and a chromosomal aberration test using human lymphocytes resulted in negatives as in vitro tests (SIDS (2004)).

- - In a two-generation reproductive toxicity test by drinking water administration to rats (OECD TG 416, GLP), no adverse effects on fertility and reproductive organs/tissue in each generation at a dose (5000 ppm) where general toxicity (decreases in water and food consumption, decreased body weight gain) was observed in parent animals, and no adverse developmental effects except decreased body weight, changes in organ weights, and delayed sexual maturation in fetuses at the same dose were reported. Besides, in an oral administration test to pregnant rabbits during an organogenetic period (OECD TG 414, GLP), no adverse developmental effects including teratogenicity except increased skeletal variations in fetuses (an increase in 13th ribs) at a dose (540 mg/kg/day) where maternal animals showed general toxicity (decreases in food consumption and body weight gain) were observed (SIDS (2004)).
From the above, it was classified as "Not classified" because neither adverse effects on sexual function and fertility nor fetal development was observed.

A death of a male in a 1000 mg/kg group and toxic signs of apathy, staggering gait, atonia, and absence of pain and corneal reflex after oral administration of 1000 and 2000 mg/kg (5 each of male and female rats per each dose) to rats were described.
And the disappearance of these signs on the day of administration or later was reported. (SIDS (2004))
