- - In the oral reproduction/developmental toxicity screening test in rats (OECD TG421, GLP-compliant), as for the toxic effects of parent animals, temporary salivation after administration in males and thickening of the mucosa and diffuse squamous hyperplasia in the forestomach in both sexes were observed at 60 mg/kg group. As for the reproductive effects, no effects related to the treatment were observed in the estrous cycle, copulation or fertility. In addition, no effects related to the treatment were observed in gestation length, parturition, and lactation. In the acute oral toxicity study in rats (OECD TG 401, GLP-compliant), three rats in 600 mg/kg group in both sexes and all rats in 1200 mg/kg group died from 6 hours after dosing until day 3. The survivors of 600 mg/kg group in both sexes revealed suppression of body weight gain. As clinical symptoms, the signs of toxicity appeared at one to six hours after dosing in males and females of 600 mg/kg group included bradypnea, hypothermia, lateral position, decreased locomotor activity, and staining of fur around the external urethral orifice, etc. As necropsy findings, dark reddish spots on the mucosa of glandular stomach or forestomach were observed in female decedents in 600 and 1200 mg/kg groups, however, no macroscopic changes were noted in the survivors in both sexes ("Toxicity Testing Reports of Environmental Chemicals" (Chemicals Investigation Promoting Council) (Access on Jan. 2011)). The above findings were the effects observed within the range of dose levels corresponding to Category 2 of the guidance values, but it was difficult to specify the target organ(s) based merely on the information. As for the gastric effects, although the irritating potency of the substance was unknown, because the possibility of its local irritant effects or for the physical effects attributed to repeated oral doses through the gastric tube could not be denied, they were not evidenced to classify. On the contrary, higher value of the liver weight and perilobular fatty change were observed at 300 - 400 mg/kg/day (converted value as that of 90-day study: 93.3 - 124.4 mg/kg/day, near the upper limit dose level of the range of the guidance values) ("Toxicity Testing Reports of Environmental Chemicals" (Chemicals Investigation Promoting Council) (Access on Jan. 2011)), however, the toxicological significance of these findings could not be judged from the only data provided without any further data in the higher dose levels.
