In a test by a 4-hour occlusive application of 0.5 mL of the undiluted test substance to the skin of six rabbits, the average scores at 24, 48, 72 hours after removal of the patch were 2.0, 2.5, 2.5 for erythema and 3.3, 2.7, 2.3 for edema (SID (2003)). Sluggishness, prostration, moribund appearance, lacrimation, unsteady gait, and tremors were observed in the groups of 1,000 mg/kg or above in an acute oral toxicity test with rats (LD50: 1,640 mg/kg) (SIDS (2003)), and apathy, ataxia, and closed eyes in the 1,000 mg/kg dose group, and apathy, ataxia, prostration, reduced or no reflexes, and labored breathing in the groups of 2,150 mg/kg or above were found in another acute oral toxicity test with rats (LD50 value: 3740 mg/kg) (SIDS (2003)). From the above, after oral administration to rats, neurobehavioral symptoms were seen at 1,000 mg/kg corresponding to the guidance value range for Category 2, and narcosis-like actions such as apathy and reduced reflexes were observed in the groups of 2,150 mg/kg or above. Therefore, it was classified in Category 2 (nervous system), Category 3 (narcotic effects). And it was classified in Category 3 (respiratory tract irritation) because it is described that there were no deaths in an inhalation test by exposure to vapour in rats (dose: 6.1 mg/L/4 hours), but irritation to the respiratory tract was found (SIDS (2003)). In a 2-week repeated inhalation exposure test with rats (vapour: 6 hours/day, total 9-time exposure) (GLP-compliant), findings were edema in the nasal cavity and squamous metaplasia in the respiratory mucosa in the low dose group (480 mg/m3, converted guidance value: 0.075 mg/L), mild to moderate rhinitis, cell degeneration or atrophy of the olfactory epithelium, squamous metaplasia in the epithelium of the nasal cavity and to a lesser degree in the larynx and trachea in the middle dose group (4,660 mg/m3, converted guidance value: 0.725 mg/L), and signs of respiratory distress, respiratory tract injury by histological examination, and deaths of all animals (30/30) in the high dose group (20,640 mg/m3, converted guidance value: 3.21 mg/L) (SIDS (2003)).
