There is a report of a positive intraperitoneal mouse bone marrow chromosomal aberration test (in vivo somatic cell mutagenicity test) (NTP DB (access on Oct. 2009)). From in vitro mutagenicity tests, there are reports of positive Ames tests, Chinese hamster ovary cell SCE tests and chromosomal aberration tests (NTP DB (access on Oct. 2009)), human cell chromosomal aberration tests and DNA binding tests (IARC vol. 40 (1986)). Based on the classification of "Group 2A" in IARC (IARC suppl. 7 (1987)), the substance was classified into Category 1B. In animal tests, skin tumours (mainly papillomas) was observed when mice received topical application and simulated solar irradiation for 37 weeks (IARC vol. 40 (1986)) and squamous-cell carcinomas invading the dermis and cartilage was observed when mice received topical application plus UV (wavelengths <340 nm) irradiation for 23 weeks (IARC vol. 40 (1986)). Epidemiologically, it was reported that "keratomas" or "epitheliomas" of the skin were found in exposed workers in the bergamot oil (containing the substance as an ingredient) production industry. Based on a report that there was a dose-dependent increase in fetal abnormalities at dose levels at which there was maternal toxicity (decreased weight gain; only 50% of dam gave birth to living pups) when rabbits were administered orally during organogenesis period (IARC vol. 40 (1986)), the substance was classified into Category 2. There is a report that hepatomegaly, necrosis and hepatic inflammation occurred at a dose level of 19 mg/kg (90-day equivalence) (administration for 28 days) in a dog oral test (IARC vol. 40 (1986)).
