In 87-week oral administration tests using rats, 92% of treated animals developed liver cancer 4-35 weeks after administration (IARC vol. 1, 1972; HSDB, 2003). Similarly, in 9-month oral administration tests, malignant hepatoma was observed at concentrations of 1500 and 2000 mg/kg (DFGOT vol. 4, 1992). In addition, in 52-week oral administration tests using mice, frequent cases of hepatoma (auramine-treated/control = 7/0) and lymphoma (auramine-treated/control = 11/5) were observed relative to control (IARC vol. 1, 1972; DFGOT vol. 4, 1992; HSDB, 2003). Furthermore, according to existing classifications for carcinogenicity, the substance is rated as Group 2B by the Japan Society for Occupational Health, Group 2B (technical-grade) by the IARC, and 3 by the EU. - - In oral administration tests using rats, a substance containing a low concentration of auramine (the subsntance consisted of 87% dyestuff with remainder including NaCl, Na2SO4, and traces of Michler's ketone) was applied at either 400 ppm (90-day conversion: 60 mg/kg/day), a concentration that falls under Category 2, for four weeks, or at 200 ppm (10 mg/kg/day) for 90 days or for two years- no toxic effects were observed in any of the tests (DFGOT vol. 4, 1992). On the other hand, in 9-month oral administration tests using rats that received 300, 500, 1000, 1500, or 2000 mg/kg/day, which are above Category 2 guidance doses, the following changes were noted at the dose of 2000 mg/kg: cirrhosis, adenofibrosis, bile duct proliferation, and mataplastic changes of the liver. Similar changes were also detected after the application of 1500 mg/kg. Foci of modified hepatocytes were noted after 3 months at doses of 1500 and 2000 mg/kg, after 6 months at 500 mg/kg, and after 9 months at 300 mg/kg (DFGOT Vol. 4, 1992). Overall, since no tests have been conducted at doses within the range of guidance values, the substance was classified into the "Classification not possible" category.
