It is reported that oral administration to rats on gestational days 0-20 caused cataract in 38% of the offspring (HSDB (2003), Teratogenic (12th, 2007), corresponding to List 2). And as for humans, eye defects were observed in an infant after treatment by this substance on the mother during gestation, and medication with this substance was considered to be a risk factor for limb defects among 108 cases analyzed. Furthermore, possible associations were found with cardiovascular anomalies, Down syndrome, and polydactyly, which were seen in 537 mother-child pairs exposed during the first trimester (Birth Defects (3rd, 2000), corresponding to List 2). From the above knowledge, general toxicity in maternal animals was not described in the animal test, and all the descriptions in humans were from List 2, therefore it was classified in Category 2. | FY2012 | GHS Classification Guidance by the Japanese Government (July, 2010) |
8 | Specific target organ toxicity - Single exposure | Classification not possible |

- | - | Data are lacking. Besides, it is reported that after administration of 80-grain tablets (about 5.2 g) to a dog, it showed increased pulse rate and behavior indicating loss of vision, no light reflex and dilated pupils were observed, and it died after 12 hours (HSDB (2003)), but it cannot be used for the rationale for classification because the details are unknown. And sodium L-thyroxine (CAS 55-03-8), the sodium salt of the substance, has been used as a medicine, and as adverse effects, there are descriptions of angina and hepatic function disorder as well as circulatory system symptoms such as palpitation, arrhythmia, and increased pulse rate and neurological symptoms such as dizziness, insomnia, tremor, and irritability (Ethical Pharmaceuticals (2010), corresponding to List 1). | FY2012 | GHS Classification Guidance by the Japanese Government (July, 2010) |
9 | Specific target organ toxicity - Repeated exposure | Classification not possible |
