From RD50 (concentrations depressing the respiratory rate to 50%) of 541 ppm for this substance in an inhalation exposure test in mice and description of a respiratory irritant (HSDB (2008)), it was classified in Category 3 (Respiratory tract irritation).
In addition, from an EC50 of 770 ppm for rotarod performance decrements (PATTY (5th, 2001)) and EC50 of 850 ppm for decrements in pain sensitivity (PATTY (5th, 2001)) in inhalation exposure to rats and the information that the acute toxicity of trimethylbenzene is due mainly to effects on the central nervous system (PATTY (5th, 2001)), it was classified in Category 3 (narcotic effects).
Besides, it is reported that neither irritation nor central nervous symptoms were observed in a test exposed 25 ppm to 10 subjects for 2 hours (PATTY (5th, 2001)).

- - A 28-day repeated oral administration test in rats reported a decreased weight gain, changes in hematological and biochemical examinations such as prolonged prothrombin time and high total protein, increases in liver and kidney weights in a high dose group at 1000 mg/kg (converted to a 90-day equivalent value: 311 mg/kg/day), and hepatocyte swelling and hyaline droplets in the kidney as pathological signs at doses of 300 mg/kg (converted to a 90-day equivalent value: 93.3 mg/kg/day) or higher (Report on Toxicity Tests of Chemical Substances, MHLW (2011)). However, the changes in the liver were adaptive changes and the changes in kidney were nephropathy specific for male rats, therefore, it corresponds to "Not classified" in an oral route due to no serious adverse effects to human.
On the other hand, a 3-month repeated administration test by inhalation exposures to vapours in rats reported an increase in liver weight and changes in hematological parameters such as a decrease in red blood cells in a high dose group (1.23 mg/L) that was above a range of Guidance values, and an increased number of goblet cells and lesions in lung including interstitial lung parenchyma infiltration in both sexes in a high dose group (HSDB (2008)). However, the classification is not possible because effects around the upper limit in a range of Guidance values (1 mg/L) are unknown.
On the other hand, the classification is not possible in dermal administration due to no data.
From the above, it was classified as "classification not possible" in specific target organ toxicity (repeated exposures).

If chronic toxicity data are used, then it is classified in Category 2 due to being not rapidly degradable (a 14-day degradation rate by BOD: 0% (Biodegradation and Bioconcentration Results of Existing Chemical Substances under the Chemical Substances Control Law, METI, 1980)) and 48-hour NOErC = 0.38 mg/L for algae (Raphidocelis subcapitata) (Results of Aquatic Toxicity Tests of Chemicals conducted by Ministry of the Environment in Japan (Ministry of the Environment, 2012)).
If acute toxicity data are used for a trophic level for which chronic toxicity data are not obtained (crustacea, fish), then it is classified in Category 2 due to being not rapidly degradable and 48-hour EC50 = 2.7 mg/L for crustacea (Daphnia magna) (Results of Aquatic Toxicity Tests of Chemicals conducted by Ministry of the Environment in Japan (Ministry of the Environment, 2012)).
From the above results, it was classified in Category 2. The classification result was revised from the previous classification by changing how to classify it in chronic toxicity and using new information.
