An rat inhalation test in exposures to vapours at 10 to 160 ppm for 4 weeks or at 26 to 224 ppm for 3 months reported alveolar macrophage accumulation accompanied by an increase in lung weights and interstitial inflammation at 4 weeks at a maximal concentration of 160 ppm (corresponds to 50 ppm = 0.745 mg/L for 90 days) as well as at 3 months at a maximal concentration of 224 ppm (3.40 mg/L), and histopathological evidence indicated the primary target organ following inhalation exposure was the lung (HSDB (2006)).
Because the concentration that affected the lung in the 4-week inhalation test corresponds to a range of Category 2 in Guidance values, it was classified in Category 2 (lung).
Besides, an increase in liver weights and changes in serum biochemical parameters (an increase in gamma-GT) were observed at the same dose, but they were not used as bases for classification due to lack of histopathological evidence in the liver.
