In a rat and mouse oral (gavage) 14-week test, forestomach squamous epithelial hyperplasia was observed at dose levels within Category 2 (male rats: >12 mg/kg, female rats: 25 and 50 mg/kg, male mice: 32 and 62.5 mg/kg, female mice: 16, 32 and 62.5 mg/kg) and forestomach necrosis and hemorrhage were present in most rats (100 mg/kg). Male mice had significantly increased incidences of glandular stomach hemorrhage (62.5 mg/kg). Bone marrow hyperplasia, depletion in the lymph nodes and necrosis of the thymus were observed (rats: 100 mg/kg, mice: 62.5 mg/kg). Based on this information (NTP TR48 (2006)), the substance was classified into Category 2 (bone marrow, lymph system). Although adverse effects on the stomach are apparent, they are considered to be effects by site of contact due to oral (gavage) administration of an irritant and are not classified in specific target organ toxicity.
