5 | Germ cell mutagenicity | Classification not possible |
- |
- | - | Regarding in vitro mutagenicity tests using 3,3'-dimethylbenzidine dihydrochloride (CAS: 612-82-8), a reverse mutation test using Salmonella typhimurium and a chromosomal aberration test using CHO cells both gave positive results (NTP DB, accessed in November 2007). However, since no appropriate in vivo test results are available, classification is not possible. 7 | Reproductive toxicity | Classification not possible |
- |
- | - | In male rats given water containing 3,3'-dimethylbenzidine dihydrochloride (CAS: 612-82-8) an increased number of immature sperm was noted in the epididymis (Initial Environmental Risk Assessment of Chemicals (Ministry of the Environment), vol. 4, 2005). However, since we are unable to determine if administration in fact impairs fertility based on the report, classification is not possible. See also the classification of 3,3'-dimethylbenzidine (CAS: 119-93-7) (NITE GHS classification ID 148). |

In humans, inhalation of small quantities of 3,3'-dimethylbenzidine dihydrochloride (CAS: 612-82-8) induced sneezing followed by upper respiratory tract irritation (Initial Environmental Risk Assessment of Chemicals (Ministry of the Environment), vol. 4, 2005). Thus, the substance was classified into Category 3 for respiratory tract irritation. See also the classification of 3,3'-dimethylbenzidine (CAS: 119-93-7) (NITE GHS classification ID 148). |

9 | Specific target organ toxicity - Repeated exposure | Category 1 (liver, kidney, lymph node, pancreas, thyroid, bone marrow) |
Danger |
H372 | P260 P264 P270 P314 P501 |
In the experiment in which rats were administered with 3,3'-dimethylbenzidine dihydrochloride (CAS: 612-82-8) in drinking water for 13 weeks, the following effects were observed in treated rats: hepatocellular necrosis, accumulation of bile pigments, increased severity of nephtopathy, lymphocytic atrophy of the thymus, lymphocytic atrophy of the mandibular and mesenteric lymph nodes, focal acinar cell degeneration in the pancreas, decreases in serum triiodothyronine (T3) values, decreases in thyroxin (T4) concentrations, and atrophy of the bone marrow in high dose groups (NTP TR 390, 1991). Thus, we determined that the liver, kidney, lymph node, pancreas, thyroid gland, and bone marrow are target organs. All symptoms were seen at doses that fall under Category 1. Therefore, 3,3'-dimethylbenzidine dihydrochloride was classified into Category 1 (liver, kidney, lymph node, pancreas, thyroid gland, and bone marrow) See also the classification of 3,3'-dimethylbenzidine (CAS: 119-93-7) (NITE GHS classification ID 148). |
