Two middle aged men developed severe hepatic injury just after cleaning a tank at a plant producing dichloropropanol and died from hepatic failure 4 and 11 days, respectively, following exposure. Liver specimens taken at autopsy showed submassive hepatic necrosis (HSDB (2003)). This information is reported in a document in List 2. The substance was classified into Category 2 (liver). In a rat test by oral administration for 91 to 92 days, myocardial degeneration was observed at the 35 and 100 mg/kg/day dose levels. Additionally, hypertrophy, karyomegaly and bile duct proliferations in the liver were observed at the 100 mg/kg/day dose level and serum enzyme changes were seen at the 35 and 100 mg/kg/day dose levels (IRIS (2004)). Since these effects on the heart and liver are observed at dose levels within the guidance value range for Category 2,the substance was classified into Category 2 (heart, liver).
