In a continuous breeding test with mice by diet administration during a 7-day pre-copulation period and a 98-day copulation period, a dose-dependent impairment of both fertility and reproduction occurred, and almost all reproductive parameters were adversely affected. As a result of cross-over mating, effects on the female reproductive system were shown (NTP TR442 (1994), NTIS PB90-253766 (1990)). Because the above effects were also observed in the dose group that did not show reduced weight gain as general toxicity, it was classified in Category 1B. In an acute oral toxicity test with rats, infiltration of red blood cells in the liver and myeloid metaplasia of the red pulp of the spleen are reported in the group given 1,960 mg/kg (NTP TR442 (1994)). Furthermore, similar findings were obtained after oral administration to mice (IUCLID (2000)). And it is reported that methemoglobin formation and hematopoietic toxicity are found after the administration of aromatic nitro compounds, and this toxicity is often more severe in rats than in mice (NTP TR442 (1994)).
