2 | Skin corrosion/irritation | Not classified |
- |
- | - | It was classified as "Not classified" because it is reported that there was no irritation in a test with rabbits (OECD TG 404) (IUCLID (2000)). | FY2013 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.0)) |

5 | Germ cell mutagenicity | Classification not possible |
- |
- | - | Classification not possible due to lack of data. As for in vivo, it was negative in a micronucleus test with mouse peripheral blood erythrocytes and a chromosomal aberration test with bone marrow cells of Chinese hamsters (NTP TR442 (1994), IUCLID (2000), NTP DB (Access on September 2013)), and as for in vitro, it was positive in a bacterial reverse mutation test, and a chromosomal aberration test with cultured mammalian cells (NTP TR442 (1994), IUCLID (2000), NTP DB (Access on September 2013)), and negative in an hgprt gene mutation test (IUCLID (2000)). | FY2013 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.0)) |
6 | Carcinogenicity | Classification not possible |
- |
- | - | Classification not possible due to lack of data. Besides, it is reported that in 2-year diet administration tests with rats and mice, there was no evidence of carcinogenicity in male rats, increases in the incidences of clitoral gland adenoma and clitoral gland adenoma or carcinoma (combined) were observed in female rats, and there was no evidence of carcinogenicity in both sex of mice (NTP TR442 (1994)). | FY2013 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.0)) |
7 | Reproductive toxicity | Category 1B |
Danger |
H360 | P308+P313 P201 P202 P280 P405 P501 |
In a continuous breeding test with mice by diet administration during a 7-day pre-copulation period and a 98-day copulation period, a dose-dependent impairment of both fertility and reproduction occurred, and almost all reproductive parameters were adversely affected. As a result of cross-over mating, effects on the female reproductive system were shown (NTP TR442 (1994), NTIS PB90-253766 (1990)). Because the above effects were also observed in the dose group that did not show reduced weight gain as general toxicity, it was classified in Category 1B. | FY2013 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.0)) |
8 | Specific target organ toxicity - Single exposure | Category 2 (blood system) |
Warning |
H371 | P308+P311 P260 P264 P270 P405 P501 |
In an acute oral toxicity test with rats, infiltration of red blood cells in the liver and myeloid metaplasia of the red pulp of the spleen are reported in the group given 1,960 mg/kg (NTP TR442 (1994)). Furthermore, similar findings were obtained after oral administration to mice (IUCLID (2000)). And it is reported that methemoglobin formation and hematopoietic toxicity are found after the administration of aromatic nitro compounds, and this toxicity is often more severe in rats than in mice (NTP TR442 (1994)). It was classified in Category 2 (blood system) based on the above knowledge. | FY2013 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.0)) |
9 | Specific target organ toxicity - Repeated exposure | Classification not possible |
- |
- | - | In 13-week diet administration tests with rats and mice, no toxicity effects were observed even after exposure to the doses within the guidance value range for Category 2. However, in rats, effects on the spleen (congestion, deposition of pigment (hemosiderin), increased weights) and kidney (deposition of pigment (hemosiderin), increased weights) were found from the high dose of 2,500 ppm (170 mg/kg/day) corresponding to "Not classified," and effects on the blood system (increases in methemoglobin, Heinz bodies, and reticulocyte counts, decreases in erythrocyte counts, hemoglobin, and hematocrit values) and testis (atrophy of the seminiferous tubules) were also seen at 10,000 ppm (660-680 mg/kg/day). And in 2-year diet administration tests, no non-neoplastic lesions were observed in rats at up to 2,500 ppm (100-125 mg/kg/day) and in mice at up to 5,000 ppm (675-905 mg/kg/day) (NTP TR442 (1994)). Therefore, it corresponds to "Not classified" in the oral route. However, because of no toxicity information on the other routes, the classification is not possible due to lack of data. | FY2013 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.0)) |

11 | Hazardous to the aquatic environment Short term (Acute) | Not classified |
- |
- | - | It was classified as "Not classified" from 96-hour LC50 > 500 mg/L for fish (Danio rerio) (IUCLID, 2000). | FY2013 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.0)) |
