There are positive results from in vivo somatic cell mutagenicity tests including bone marrow or peripheral blood micronucleus tests by oral gavage to mice (NTP DB (2004)), bone marrow chromosomal aberration test by intraperitoneal injection to mice (NTP DB (1983)) and erythrocyte micronucleus test by oral gavage and intraperitoneal injection to 2 mouse strains (HSDB (2000)). In addition, there is a report that the frequency of dominant lethal mutations increases with higher dose levels in mice (in vivo heritable germ cell mutagenicity test) (HSDB (2000)). Based on these results, the substance was classified into Category 1B. As relevant information, the substance is classified into "R" in NTP (NTP RoC (11th, 1983)). In a test where male rats administered by gavage for 5 days and mated to untreated females, effects on male fertility index and reduction in number of implants are reported (RTECS (2008): TXAPA9 Toxicology and Applied Pharmacology. 70 (1983)). Information on resorbed implants, fetal death and growth restriction are reported separately (RTECS (2008): JTSCDR Journal of Toxicological Sciences. 25 (2000)). In a similar test in mice, effects on male fertility index and death of offspring (RTECS (2008): JACTDZ Journal of the American College of Toxicology. 2 (2) (1983)) were reported. In addition, there are report that intraperitoneal injection to rats caused defect of limb and head (Birth Defects (3rd, 2000)) and that the substance produced subfertility and larger doses caused complete sterility in mice (IARC vol. 7 (1974)). It was also reported that intraperitoneal injection to pregnant mice induced high frequencies of midgestation and late fetal deaths, and of malformations among some of the surviving fetuses (HSDB (2008)). The data for oral route are on the document in List 3 and the details are unknown, and other data are results by intraperitoneal injection.
