---|---|---|---|---|---|---|---|---|
1 | Acute toxicity (Oral) | Category 4 |
Warning |
H302 | P301+P312 P264 P270 P330 P501 |
It was classified in Category 4 based on LD50 values of 1709 mg/kg for males and 1691 mg/kg for females for rats (OECD TG 401, GLP) (JECDB (Access on Sep. 2012)). As for health hazards, refer to 4-ethylphenol (p-ethylphenol, CAS 123-07-9), the isomer. | FY2012 | GHS Classification Guidance by the Japanese Government (July, 2010) |

4 | Skin sensitization | Classification not possible |
- |
- | - | No data available. | FY2012 | GHS Classification Guidance by the Japanese Government (July, 2010) |
5 | Germ cell mutagenicity | Classification not possible |
- |
- | - | The classification is not possible due to no in vivo test data. Besides, as for in vitro tests, it is reported that it was negative in an Ames test (JECDB (2000)) and positive in a chromosomal aberration test with CHL/IU cells (JECDB (2000)). | FY2012 | GHS Classification Guidance by the Japanese Government (July, 2010) |
6 | Carcinogenicity | Classification not possible |
- |
- | - | No data available. | FY2012 | GHS Classification Guidance by the Japanese Government (July, 2010) |
7 | Reproductive toxicity | Classification not possible |
- |
- | - | Reproductive information is limited to the report that a significant decrease in body weights and a significant increase in relative liver weights (at the end of the treatment period) were observed at 300 mg/kg/day in a test in which neonatal rats were orally dosed on postnatal days 4-21, but toxicological significance was unknown because there were no histological findings or changes in blood biochemical parameters (HPVIS (2008)). Therefore, it was classified as "Classification not possible" due to lack of data. | FY2012 | GHS Classification Guidance by the Japanese Government (July, 2010) |
8 | Specific target organ toxicity - Single exposure | Category 2 (systemic) |
Warning |
H371 | P308+P311 P260 P264 P270 P405 P501 |
In an acute oral administration toxicity test with rats (OECD TG 401, GLP), deaths occurred within about one hour to one day after the administration in the groups at or above 1,700 mg/kg, staggering gait, prone position, lateral position, etc. were observed in the groups at or above 1,450 mg/kg, and on necropsy, impairing changes such as edematous changes, brown patches, or white discoloration were found in the forestomach or glandular stomach in the dead males and females (JECDB (Access on Sep. 2012)). From the above, symptoms were seen at doses corresponding to the guidance value range for Category 2, but it was hard to specify the target organ. Therefore, it was classified in Category 2 (systemic toxicity). | FY2012 | GHS Classification Guidance by the Japanese Government (July, 2010) |
9 | Specific target organ toxicity - Repeated exposure | Classification not possible |
- |
- | - | In a 28-day repeated oral administration test with rats (doses: 0, 100, 300, 1,000 mg/kg/day, Guidelines specified by the Chemical Substances Control Law, GLP), symptoms such as lateral or prone position and staggering gait were sporadically observed, and increased GPT, increased liver weights, thickening and hyperplasia of the squamous epithelium in the forestomach, etc. were found at 1,000 mg/kg (converted guidance value: 311 mg/kg/day), and the NOEL was reported to be 300 mg/kg/day (converted guidance value: 93.3 mg/kg/day) (JECDB (Access on Sep. 2012)). Therefore, it probably corresponds to "Not classified" in the oral route. However, because effects in the other routes (inhalation, dermal) are unknown due to no data, it was classified as "Classification not possible" due to lack of data for specific target organ toxicity (repeated exposure). | FY2012 | GHS Classification Guidance by the Japanese Government (July, 2010) |
