In the combined repeated dose toxicity study with the reproduction/developmental toxicity screening test by oral administration in rats (OECD TG 422), in addition to low or lower tendency of number of estrous, corpora lutea of pregnancy, number of implantation and implantation index, low or lower tendency of total pups born, live pups born, delivery index, birth index, live pups on day 4 of lactation were also observed at high dose group (720 mg/kg) that induce one of 12 death and low body weights in dams ("Toxicity Testing Reports of Environmental Chemicals"(Chemicals Investigation Promoting Council) (Access on Jan. 2011)). In the acute oral toxicity study in rats (OECD TG 423), three of six animals died on day 1 after administration at 2000 mg/kg, and in addition to suppression of body weight gain signs of toxicity observed included tremors, clonic convulsions, staining around the lower abdomen or diarrhea. However, no macroscopic abnormalities were noted in both decedents and survivors at necropsy. Furthermore, no abnormalities were detected in clinical signs, macroscopic observation and the other at 300 mg/kg ("Toxicity Testing Reports of Environmental Chemicals" (Chemicals Investigation Promoting Council) (Access on Jan. 2011)). Based on the signs of tremors and clonic convulsions observed at 2000 mg/kg, equivalent to Category 2 of the guidance values, it was reported that respiratory paralysis or abnormality in central nervous system was considered as the cause of death ("Toxicity Testing Reports of Environmental Chemicals" (Chemicals Investigation Promoting Council) (Access on Jan. 2011)). In the combined repeated dose toxicity study with the reproduction/developmental toxicity screening test in rats (OECD TG 422), minimal to moderate centrilobular swelling of hepatocytes and minimal or slight basophilic changes of hepatocytes in the liver were observed in the groups at 45 mg/kg/day (converted value as that of 90-day study: 14 mg/kg/day) or higher, and minimal hyaline droplet and degeneration in the tubular epithelium in the kidneys were observed in the groups at 180 mg/kg/day (converted value as that of 90-day study: 56 mg/kg/day) or higher. At the end of the recovery period, centrilobular swelling of hepatocytes in the liver and degeneration of tubular epithelium in the kidneys still remained in the group at 720 mg/kg/day (converted value as that of 90-day study: 224 mg/kg/day) ("Toxicity Testing Reports of Environmental Chemicals" (Chemicals Investigation Promoting Council) (Access on Jan. 2011)). The changes in the liver were observed over the dose level corresponding to Category 2 of the guidance values, and they were considered to occur hepatic dysfunction accompanied by swelling of hepatocytes, based on the findings such as higher values in gamma-GTP and total bilirubin in blood biochemistry as well as prolongation of prothrombin time and activated partial thromboplastin time and higher values in fibrinogen concentration in hematology ("Toxicity Testing Reports of Environmental Chemicals" (Chemicals Investigation Promoting Council) (Access on Jan. 2011)). Therefore, the substance was classified as Category 2 (liver). As for the changes in the kidneys, however, they were accompanied by hyaline droplet in renal tubular epithelium which was specific to male rats and thus were not evidenced to classify.
