 In an oral administration test of this substance using male mice dosed at 7.5 - 30 mg/kg/day for 43 days, decreased sperm mobility and sperm counts were observed in a dose-dependant manner (Japan Society For Occupational Health (JSOH) Vol.56 (2014), ATSDR (2012)). It is also reported that in inorganic manganese compounds, such as manganese chloride or manganese sulfate, embryotoxicity and occurrences of malformations were reported. In a test in which manganese chloride tetrahydrate was subcutaneously administered to pregnant mice (Gestation days 6 to 15), at doses in which suppression of maternal weight gain and reduction in feed intake were observed in maternal animals or even at lower doses, increased embryo resorptions, hypoplasia of the kidneys, and wavy ribs were observed in fetuses (Initial Risk Assessment Report (NITE, CERI, NEDO, 2008), CICAD 12 (1999)). Also, in a test in which manganese chloride was intravenously administered to pregnant rats during organogenesis (Gestation Days 6 to 17), fetal death, skeletal anomality, wavy ribs, bending of limbs were observed even at doses lower than that of maternal toxicity (suppression of body weight gain, implantation number reduction) was observed (Initial Risk Assessment Report (NITE, CERI, NEDO, 2008), CICAD 12 (1999)). In addition, it is also reported that increased exencephaly and embryo resorptions were observed in a single intraperitoneal administration of manganese sulfate to pregnant mice on the 8th day of gestation, and at higher doses, inhibition of implantation was observed (Initial Risk Assessment Report (NITE, CERI, NEDO, 2008)).
 Although weak as epidemiological evidence, on the basis of case reports of impotence or decreased libido observed in manganese addicts, and on the basis of embryotoxicity observed in studies using experimental animals, the Japan Society for Occupational Health classified manganese and its compounds in Group 2 as reproductive toxicity (Japan Society For Occupational Health (JSOH) Vol.56 (2014), Recommendation of Occupational Exposure Limits (2016)). Therefore, from the results of studies on this substance and inorganic manganese compounds using experimental animals, it was deemed appropriate to classify this substance in Category 1B.

 As for experimental animals, in a 63-day repeated dose toxicity study by gavage using rats, at doses of 87 mg/kg/day (converted guidance value: 60.9 mg/kg/day), which is equivalent to Category 2, glomerulosclerosis/nephritis and urolithiasis (kidney stones) were observed, and at doses of 137.2 mg/kg/day (converted guidance value: 96 mg/kg/day), which is equivalent to Category 1, testicular degeneration was observed. In the 43-day repeated dose toxicity study by gavage using mice, at doses of 4.6 mg/kg/day (converted guidance value: 2.2 mg/kg/day), which is equivalent to Category 1, decreased sperm motility and sperm counts were reported (ATSDR (2012)).
 In addition, although it is not regarding this substance, when humans are exposed to excess amount of manganese, by the oral or inhalation route, memory disorder or psychiatric symptoms were observed as acute effects, and as chronic effects, symptoms of manganese addiction similar to those of Parkinson's disease were seen, such as gait disturbance and speech disorders. Serious effects were observed especially in inhalation exposure (Initial Risk Assessment Report (NITE, CERI, NEDO, 2008)).
 As for the oral route, in cases of drinking well water containing high concentrations of manganese, and in epidemiological investigations on residents living in regions with high manganese concentration, symptoms similar to manganese addiction such as mask-like faces, muscle rigidity, tremors and psychiatric disorders were reported (Initial Risk Assessment Report (NITE, CERI, NEDO, 2008)). The influence on the neurobehavioral function by exposure to the dust of manganese dioxide, trimanganese tetraoxide, and manganese salts (sulfate, carbonate, nitrate) in plants manufacturing manganese oxides and manganese salts were reported (Initial Risk Assessment Report (NITE, CERI, NEDO, 2008)). Occurrence of neurotoxicity due to manganese addiction has been reported due to occupational exposure to manganese dioxide. In addition to deterioration in visual simple reaction time and eye-hand coordination, rigidity of the hands have been observed (SIDS (2012), Initial Risk Assessment Report (NITE, CERI, NEDO, 2008), ATSDR (2012)). Furthermore, many case reports reveal that manganese compounds, mainly manganese dioxide, produce respiratory disorders (cough, bronchitis, pneumonia, etc.) (ATSDR (2012)).
 Therefore, this substance was classified in Category 1 (genetic organs (men)), Category 2 (kidney).
