(1) For this substance (DHP), no fertility or developmental toxicity study with mammals was available, but it was proposed to classify it in Repr. 1B by the read-across method based on the data of six phthalates with a similar structure (diisobutyl phthalate (DIBP) CAS RN 84-69-5, di-n-butyl phthalate (DBP) CAS RN 84-74-2, diisopentyl phthalate (DIPP) CAS RN 605-50-5, di-n-pentyl phthalate (DPP) CAS RN 131-18-0, di-n-hexyl phthalate (DnHP) CAS RN 84-75-3, diethylhexyl phthalate (DEHP) CAS RN 117-81-7) (CLH Report (2012)).
(2) DIBP, DBP, DPP, DEHP, and DnHP were classified in Category 1B for reproductive toxicity in the GHS classification by the Japanese government (classification in FY 2018, 2013, 2008, 2014, and this fiscal year (2020) respectively). (1) It was reported that in a 13-week test with rats dosed by feeding, in a 0.05% group (the dose was increased to 1.0% (776 mg/kg/day) in week 7 and a 3.0% group (2,298 mg/kg/day) in week 12), effects on the male reproductive organs (a decrease in testis weight, atrophy of spermatogenic epithelium) were observed, and reduced body weight gains, a decrease in food consumption, respiratory distress, stiff gait, rigid tail, decreases in weight of the gonad, spleen, kidney, and adrenal gland, an increase in liver weight, eosinophilic areas, variations in the size of the nuclei, etc. in the liver were also observed (CLH Report (2012)).
(2) It was reported that in a 13-week test with dogs dosed by feeding, in a 5.0% group (the dose was 0.1% at the start and was increased to 5.0% in weeks 9-13), an increase in liver weight in males and females, and a decrease in testis weight, atrophy of seminiferous tubular epithelium, and enlarged hepatic cells in males were observed (CLH Report (2012)).
