 In 2-year carcinogenicity studies in rats and mice administered by drinking water, increased incidences of malignant mesothelioma and tumors in the oral cavity (oral mucosa and tongue), large intestine, brain, skin, and mammary gland were observed in rats. In mice, an increased incidence of tumors in the liver and Harderian gland was observed. From the above, it was concluded that there was clear evidence of carcinogenic activity in both sexes in rats and mice (NTP TR 583 (2015)). There are no classifications by other organizations, however, the substance was classified in Category 1B because tumors in multi-organs in two species of experimental animals had been observed.  As for experimental animals, in a 104-week repeated dose toxicity study using rats given by drinking water, effects on the hematopoietic system (angiectasis and hyperplasia in the bone marrow, hematopoietic cell proliferation in the liver and spleen) were reported at 250 to 1000 mg/L (converted guidance value: 11 to 57 mg/kg/day), which is within the range of Category 2 (NTP TR 583 (2015)). Also in a 105-week repeated dose toxicity study using mice given by drinking water, effects on the liver (eosinophilic focus and atypical focus of cellular alteration (i.e., eosinophilic cell focus and mixed cell focus), effects on the testis (atrophy) and effects on the epididymis (atrophy, epithelium degeneration) were reported at 250 to 500 mg/L (converted guidance value: 17 to 52 mg/kg/day), which is equivalent to Category 2 (NTP TR 583 (2015)). Moreover, in a 14-week repeated dose toxicity study using rats given by drinking water, effects on the testis (decreased testis weight and sperm motility) were reported at 72 mg/kg/day, which is equivalent to Category 2 (NTP TR 583 (2015)). Therefore, the substance was classified in Category 2 (liver, genetic organs (men), hematopoietic system).
