[Rationale for the Classification] There was no data on this substance, but based on (1) to (4), it was classified in Category 1B. [Evidence Data] (1) A mixture (mainly DIHP, containing DnHP < 25%) consisting of this substance (DIHP) and linear di-n-hexyl phthalate (DnHP: CAS RN 84-75-3) was dihexyl (branched/linear) = phthalate (DHP: CAS RN 68515-50-4) (RAC (Background Document) (2017)). (2) In the GHS classification by the Japanese Government, DnHP was classified in Category 1B based on toxicity on the male reproductive organs, effects on fertility, and developmental effects (an increase in dead fetuses, cleft palate, abnormality in the eye, a decrease in anogenital distance of male fetuses, an increase in undescended testes, etc.) as evidence (this year (2020)). DHP was classified in Repr.1B by the read-across method based on the data of six kinds of phthalates with similar structure in the EU, and 5 out of 6 kinds were classified in Category 1B in the GHS classification by the Japanese Government, and on the basis of them, it was classified in Category 1B (this year (2020)). (3) For this substance, there was no available fertility or developmental toxicity study with mammals, but it was proposed to classify it in Repr. 1B by the read-across method based on the data of phthalates with similar structure (diisobutyl phthalate (DIBP) (CAS RN 84-69-5), di-n-butyl phthalate (DBP) (CAS RN 84-74-2), diisopentyl phthalate (DIPP) (CAS RN 605-50-5), di-n-pentyl phthalate (DPP) (CAS RN 131-18-0), diethylhexyl phthalate (DEHP) (CAS RN 117-81-7), di-n-hexyl phthalate (DnHP) (CAS RN 84-75-3), 1,2-benzenedicarboxylic acid, dihexyl ester, branched and linear (DHP) (CAS RN 68515-50-4)) (EU CLP CLH (2017), Classification in EU CLP (Access on June 2020)). (4) Among the phthalates in (3) above, in addition to the substances shown in (2), DIBP, DBP, DPP, and DEHP were classified in reproductive toxicant Category 1B in the GHS classification by the Japanese Government (2018, 2013, 2008, and 2014 respectively). Furthermore, for DEHP, the additional category (effects on or via lactation) was added (Classification in 2014). For DIPP, the GHS classification by the Japanese Government has not been done to date. [Reference Data, etc.] (5) DHP and DnHP were already classified in Repr. 1B as a reproductive toxicant in the EU (Classification in EU CLP (Access on June 2020)). (6) In DPP, DnHp, and DEHP, reproductive toxicity findings (decreases in reproductive capacity and a number of viable pups, effects on the male reproductive organs (testis lesions)) were observed; and in DIBP, DBP, DPP, DnHP, and DEHP, developmental toxicity findings (a decrease in anogenital distance, degeneration of the seminiferous tubules, and damage of the testis) were observed (EU CLP CLH (2017)). |
