World Rev Pestic Cont 9:119-127, it was described in the original source that LD 50 values were 400-4,000 mg/kg), 3,400 mg/kg (HSDB (Access on August 2017), original source: Montgomery, J.H.; Agrochemical's Desk Reference 2nd ed. In a carcinogenicity test, where this substance for industrial use (p, p'-DDD: containing about 60% of this substance and 19 or more impurities) was administered by feeding for 78 weeks to rats and mice, and planned slaughtering was performed after an untreated period of 35 weeks for rats and 15 weeks for mice, there was no evidence for carcinogenicity in female rats and both sexes of mice. However, a significant increase in the combined incidence of follicular cell adenomas and follicular cell carcinomas of the thyroid was observed in male rats in the low dose (1,648 ppm) group (in the high dose (3,294 ppm) group, it was only an increased trend and was not significant), and a possibility of carcinogenicity was suggested. However, tumors of the thyroid were also observed in control male rats, so NCI concluded that a definitive interpretation of the data was not possible (NTP TR131 (1978), IRIS (1988)). On the other hand, in a 130-week test in which this substance was administered by feeding to mice, lung tumors in both sexes and liver tumors in males were observed at a dose corresponding to 42.6 mg/kg/day (IRIS (1988), ATSDR (2002)). Because there was sufficient evidence of carcinogenicity in experimental animals, and this substance is a known metabolite of DDT (dichlorodiphenyltrichloroethane: CAS RN 50-29-3), this substance was classified in B2 (Probable human carcinogen) by EPA (IRIS (1988)).
From the above, the substance was classified in Category 1B based on the animal test results and classification results by other organizations. Besides, as for carcinogenicity of the parent compound DDT, IARC announced that the classification would be changed from the previous Group 2B to Group 2A (IARC Press release No. 236 (2015)).
