[Rationale for the Classification]
Since the central nervous system depression and effects on the heart were observed in humans in (1) and (2), and effects of central nervous system depression were found at high concentrations in experimental animals in (3) and (4), it was classified in Category 1 (cardiovascular system), Category 3 (narcotic effects). By review of the information, the classification result was changed from the previous classification.

[Evidence Data]
(1) In a study in which 4 volunteers were exposed by inhalation to 5% and 6 volunteers to 10% of this substance for a maximum of 20 minutes, 3 of the 4 persons in the 5% group complained of slight light-headedness and feeling of relaxation which were considered to be effects on the central nervous system, and all 6 persons in the 10% group complained of confusion, unsteadiness and giddiness. The authors therefore concluded that these were the first signs of narcotic effects. In the results from the electrocardiograms, a lowering or flattening of the T-wave was observed in three persons, one of whom showed bigeminy and atrioventricular dissociation (DFGOT vol.6 (1993)).
(2) In a test in which volunteers were exposed by inhalation to 10% and 15% of this substance, a feeling of impending unconsciousness, paresthesia, and diminished performance were observed. In addition, at the highest concentration (16.9%), auriculoventricular dissociation and premature ventricular contractions were recorded (ACGIH (7th, 2001)).
(3) In a test in which monkeys were exposed by inhalation to 20%-25% of this substance, performance decrements in operant conditioning were observed (ACGIH (7th, 2001)).
(4) In a test in which mice were exposed by inhalation to this substance at 800,000 ppm (with added oxygen of 15-20%) (converted 4-hour equivalent value: 565,700 ppm, exceeding the range of Category 2) for 2 hours, activity reduced, respiratory distress, anxiety, sleepiness, slight tremors, balance and postural disorders were observed, but there were no death cases (DFGOT vol.6 (1993)).

[Evidence Data]
(1) When rats and guinea pigs were exposed by inhalation to an average of 23,000 ppm (exceeding the range of Category 2) for 18 weeks, no signs of intoxication were observed (ACGIH (7th, 2001)).
(2) When rats, guinea pigs, dogs and cats were exposed by inhalation at a concentration of 20% for 4 weeks (3.5 hours/day, 5 days/week) (converted guidance value: 2,590 ppm, exceeding the range of Category 2), there were no changes in blood or urine parameters, and also no histopathological changes were observed other than irritant changes in the trachea (increased mucous secretion) in dogs and guinea pigs (DFGOT vol.6 (1993)).
