- - In fertility tests in rats, after inhalation exposure for 69 days (male) or 21 days (female), the males and females were mated to untreated females and males, respectively. Although a slight decreased body weight was observed in the high dose group, there were no exposure-related effects on fertility, pregnancy rates or fetal development (ECETOC JACC 53 (2007)). In a developmental toxicity test by oral administration to rats during organogenesis period, no significant differences from controls were observed for any fetal developmental parameter at a dose in which maternal general toxicity (reduction in body weight gain) was noted and no adverse effects were reported on fetal development including teratogenicity (ECETOC JACC 53 (2007)). There are many human case reports of accidental exposure (mainly dermal). Symptoms of exposure included lost of consciousness, comatose, dyspnea, and tonic-clonic convulsions. In some cases, exposed individuals died (ACGIH (2001)). Since lost of consciousness and tonic-clonic convulsions were reported and there is a report that acute exposure may cause CNS stimulation (HSDB (2005)), the substance was classified into Category 1 (central nervous system). When 5 mg/animal was administered orally to rats twice a week for 3 - 8 months (the dose level is estimated to be 12.5 mg/kg for a body weight of 400 g and the daily dose level is calculated to be approximately 4.1 mg/kg/day because of twice a week administration), decreases of total protein, glycoprotein and A/G ratio, pyknosis, anisokaryosis and fatty degeneration of hepatocyte as well as kidney lesions were found (ACGIH (2001), PATTY (5th, 2001)). Based on this report, the substance was classified into Category 1 (liver, kidney). In addition, similar findings were found in livers and kidneys in a rat test by inhalation exposure at concentration of 102 mg/L twice a week for 3 - 8 months ACGIH (2001)). Results of the inhalation test supported the results of the oral exposure test. There were no significant toxic effects in 4-week and 14-week inhalation tests in rats (highest concentration: approximately 0.21 mg/L) (CICAD 61 (2004)).
