5 | Germ cell mutagenicity | Not classified |
- |
- | - | Three dominant lethal tests in male mice by inhalation (in vivo heritable germ cell mutagenicity tests) were reported. The result of the first test showing positive effect (EHC 139 (1992)) is doubtful due to the low number of mated males and especially low incidence of fetal death in the control group. The second test showing positive effect (EHC 139 (1992)) is difficult to evaluate due to lack of a dose relationship. Finally, the negative result of the third test is employed as a conclusion (ECETOC JACC (1990)). Additionally, a negative result was observed in an inhalation rat bone marrow chromosome aberration test (in vivo somatic cell mutagenicity tests) (EHC 139 (1992)). Based on all data, the substance was classified as "Not classified". As relevant information, from in vitro mutagenicity tests, there are reports of negative results for an Ames test and a BHK 21 cell transformation assay. |

6 | Carcinogenicity | Not classified |
- |
- | - | Based on the classification of "Group 3" in IARC (IARC 71 (1999), the substance was classified as "Not classified". There is a report that the incidences of uterine carcinomas and testicular interstitial-cell neoplasms were significantly increased in a rat test that was terminated at 73-week after 52-week oral administration (IARC 41 (1986)). | FY2009 | GHS Classification Guidance by the Japanese Government (March, 2009) |
7 | Reproductive toxicity | Category 1B |
Danger |
H360 | P308+P313 P201 P202 P280 P405 P501 |
In a dominant lethal test in male mice by inhalation, exposure-related decreases in the proportion of pregnant females were observed, histopathological evidence of degeneration of spermatogenic cells was observed and reduced male fertility was observed (EHC 139 (1992)). Additionally, in a rat developmental toxicity test by inhalation during organogenesis period, the ratios of females with total resorptions were increased in a dose-dependent manner and treatment-related increases in the incidence of runts, delayed ossification and hydronephrosis were also observed at doses at which evidence of toxicity in parental animals is not manifested (EHC 139 (1992)). Therefore, the substance was classified into Category 1B. | FY2009 | GHS Classification Guidance by the Japanese Government (March, 2009) |
8 | Specific target organ toxicity - Single exposure | Category 3 (Narcotic effects) |
Warning |
H336 | P304+P340 P403+P233 P261 P271 P312 P405 P501 |
A narcotic effect was observed in mice, rats and dogs by inhalation (ECETOC JACC (1990)). Inhalation of high concentrations (738,000 mg/m3 and above) induced convulsions followed by death (EHC 139 (1992)), but recovery from nonlethal exposure is rapid (EHC 139 (1992), therefore the substance was classified into Category 3 (narcotic effects). | FY2009 | GHS Classification Guidance by the Japanese Government (March, 2009) |
9 | Specific target organ toxicity - Repeated exposure | Classification not possible |
- |
- | - | Although atrophy of thymus, spleen, testis and ovary, inflammatory changes in the nasal mucosa, emphysema, oedema, bronchitis and pneumonia in the lungs along with changes in hematology, blood clinical chemistry and organ weights were observed in a test where rats were exposed to 48,000 mg/m3 for 90 days; the dose level exceeded the guidance value range. Effects for exposure at approximately the guidance cut-off value were unclear due to lack of data. Therefore, classification was not possible. As relevant information, there is a report that no effects were observed in dogs exposed to 24,000 mg/m3 for 3 months (ECETOC JACC (1990)). | FY2009 | GHS Classification Guidance by the Japanese Government (March, 2009) |
