In a rat inhalation test, a substantial increase in circulating methemoglobin was detectable at 1000 ppm/4h, deaths occurred at 2500 ppm/4h and higher concentrations, and the methemoglobin concentrations at the time of death were equivalent to 60% to 70% of the total blood pigment (ACGIH (2001)). Additionally, it is reported that the substance cause anoxic deaths due to methemoglobinemia and results in pathologically enlargement and darkening of the spleen (ACGIH (2001)). There is a report that dogs exposure at 9600 ppm/60min exhibited Heinz body-induced hemolysis, anemia, decreased hematocrit, decreased hemoglobin, reduced red blood cell count, and clinical signs consistent with histotoxic anoxia as a result of methemoglobin formation (ACGIH (2001)). Since the major effects are induced by methemoglobin formation and are observed at dose levels within the guidance value range for Category 2, the substance was classified into Category 2 (blood system). It is reported that inhalation by rats at 100 ppm/7 hr/day (116 ppm/6 hr/day) for 19 weeks resulted in mild to moderate pathologic changes in liver and interstitial and tubular nephritis in kidneys and the characteristics of the renal injury suggested fluoride toxicosis. (ACGIH (2001)). Since the exposure level is within the guidance value range for Category 2, the substance was classified into Category 2 (liver, kidney). On the other hand, the exposure failed to cause significant effects on the spleen or on hematological parameters and there was no evidence of fluorosis in the teeth or bones (ACGIH (2001)).
