5 Germ cell mutagenicity Classification not possible
-
-
- - [Rationale for the Classification]
Classification not possible due to lack of data.

[Reference Data, etc.]
(1) There is no information on this substance.
(2) As for in vivo tests with an isomer mixture including this substance, there is a negative result in an unscheduled DNA synthesis test with rat hepatocytes (Environmental Risk Assessment for Chemical Substances Vol.4 (Ministry of the Environment, 2005)).
(3) As for in vitro tests with an isomer mixture including this substance, there are negative results in bacterial reverse mutation tests and a chromosomal aberration test with cultured mammalian cells (NTP TR433 (1994), Environmental Risk Assessment for Chemical Substances Vol.4 (Ministry of the Environment, 2005), ATSDR (2012), ACGIH (7th, 2016)).

6 Carcinogenicity Classification not possible
-
-
- - [Rationale for the Classification]
There are no classification results by domestic and international organizations. There is no available report on humans. Though there are results on isomer mixture in (1), there are no data on this substance. Therefore, the classification was not possible.

[Reference Data, etc.]
(1) It was concluded that there was no evidence of carcinogenic activity because no incidence of treatment-related tumours was observed in tests in which an isomer mixture including this substance (this substance 4%) was administered by feed to rats and mice for 2 years (NTP TR433 (1994)).

FY2019 GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1))
9 Specific target organ toxicity - Repeated exposure Classification not possible
-
-
- - [Rationale for the Classification]
Since (1) is the only information obtained for this substance, it was classified as "Classification not possible" due to lack of data.

[Reference Data, etc.]
(1) In a test in which male rats were given 100 mg/kg/day (converted guidance value: 70 mg/kg/day, within the range of Category 2) of this substance by gavage for 63 days, decreased sperm concentrations were observed (Environmental Risk Assessment for Chemical Substances Vol.4 (Ministry of the Environment, 2005)).
(2) Tri-2-Tolyl phosphate (CAS RN 78-30-8), the ortho isomer of this substance, has strong neurotoxicity called OPIDN (organophosphate-induced delayed neuropathy), and it is known that this is a toxic effect from the metabolite, saligenin cyclic phosphate. However, the m, and p-isomers do not generate such cyclic phosphates and do not cause OPIDN (Environmental Risk Assessment for Chemical Substances Vol.4 (Ministry of the Environment, 2005)).

10 Aspiration hazard Classification not possible

11 Hazardous to the aquatic environment Short term (Acute) Category 1

Warning
H400 P273
P391
P501
It was classified in Category 1 from 96-hour LC50 = 0.061 mg/L for fish (Lepomis macrochirus) (EHC 110, 1990). FY2019 GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1))
11 Hazardous to the aquatic environment Long term (Chronic) Category 1

Warning
H410 P273
P391
P501
If chronic toxicity data are used, then it is classified in Category 1 due to being rapidly degradable (a degradation rate by BOD: 100% (Biodegradation and Bioconcentration Results of Existing Chemical Substances under the Chemical Substances Control Law, 1977)), and 6-week NOEC = 0.01 mg/L for fish (Jordanella floridae) (EHC 110, 1990).
If acute toxicity data are used for a trophic level for which chronic toxicity data are not obtained, then it is classified in Category 2 due to 96-hour EC50 = 1.5 mg/L for algae (Scenedesmus pannonicus) (EHC 110, 1990) and a high bioaccumulation estimate (log Kow = 6.34 (KOWWIN)), although it is rapidly degradable (a degradation rate by BOD: 100% (Biodegradation and Bioconcentration Results of Existing Chemical Substances under the Chemical Substances Control Law, 1977)).
By drawing a comparison between the above results, it was classified in Category 1.

12 Hazardous to the ozone layer Classification not possible

- - Classification not possible due to lack of data. FY2019 GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1))
