It is described that in a 90-day diet administration test with rats, effects on the blood system (increased reticulocytes and methemoglobin, decreases in RBC counts, hematocrit, and hemoglobin), kidney (discoloration, nephropathy of the cortex and medulla) and spleen (congestion, hemosiderin deposition) were observed at the dose (2,000 ppm: corresponding to 100 mg/kg/day) of the upper limit for Category 2 (SIDS (2004)), and in a test with rats exposed to this substance (dust) by inhalation for 21 days, respiratory irritation and increased methemoglobin were found from the concentration (converted guidance value: 0.0067 mg/L/6 hours) corresponding to Category 1 (SIDS (2004)). Because the findings in the spleen in the diet administration test were thought to be secondary effects of anemia, the spleen was not taken as the target organ. Furthermore, as for respiratory irritation in the inhalation exposure test, the details of the sites and changes are unknown, including whether these were from observation of symptoms or pathological examinations. Therefore, it was judged to be inappropriate to adopt the respiratory organs as the target organs.
