[Rationale for the Classification] Based on (1) and (2), it was classified in Category 1B. [Evidence Data] (1) In a continuous breeding test with mice dosed by feeding, at doses at which no toxicity effects were observed in parental animals, reduced fertility (decreases in the number of live pups per litter and the proportion of live pups) was observed. In a histopathological examination which was conducted only for the control group and the high-dose group, a decrease in testis weight, significant effects on epididymal sperm concentration and motility, and extensive atrophy of the seminiferous tubules were observed in males in the high-dose group, and the result of a crossover mating test showed a decrease in mating capability in males (EU CLP CLH (2011), RAC Background Document (2011)). (2) In a developmental toxicity study with female rats dosed by gavage on days 6 to 20 of gestation, an increase in post-implantation embryo resorption and an increase in fetal death (by 60% compared to the control group) were observed at 750 mg/kg/day, malformations (cleft palate, eye defects) were observed at or above 500 mg/kg/day, and a significant decrease in fetal body weight, etc., increases in delayed ossification and skeletal variations were observed at 250 mg/kg/day. A decrease in anogenital distance in male fetuses was observed at or above 250 mg/kg/day and an increase in undescended testes was observed at or above 500 mg/kg/day (EU CLP CLH (2011), RAC Background Document (2011)). [Reference Data, etc.] (3) In the EU CLP classification, it was classified as Repr.1B (Classification in EU CLP (Access on October 2020)). |

8 | Specific target organ toxicity - Single exposure | Classification not possible |
- |
- | - | [Rationale for the Classification] Classification was not possible due to lack of data. [Reference Data, etc.] (1) Water-insoluble plasticisers for coating raw materials and plastics that include this substance could trigger paraesthesia in the nose and throat as well as bronchitis after the inhalation of the respective aerosols, dusts or vapors (GESTIS (Access on August 2020)). |
FY2020 | GHS Classification Guidance for the Japanese Government (FY2019 revised edition (Ver. 2.0)) |
9 | Specific target organ toxicity - Repeated exposure | Classification not possible |
- |
- | - | [Rationale for the Classification] There was information in (1) and (2), but it was not sufficient for classification. Therefore, classification was not possible due to lack of data. [Reference Data, etc.] (1) It was reported that in a 21-day oral toxicity study by administration of this substance to rats dosed at 20,000 ppm (1,824 mg/kg/day, exceeding Category 2) by feeding, an increase in liver weight, centrilobular necrosis, a decrease in glycogen, centrilobular fatty accumulation, proliferation and dilation of smooth endoplasmic reticula in the bile canaliculi, shortening of microvilli in the bile canaliculi, lipid droplets around the central veins, and an increase in peroxisomes were observed (RAC Background Document (2011)). (2) It was reported that in a 21-day oral toxicity study by administration of this substance to rats dosed at 2% (2,000 mg/kg/day, exceeding Category 2) by feeding, a decrease in serum T4 levels, an increase in thyroid activity with histopathological alterations, and fat accumulation in the liver were observed (RAC Background Document (2011)). |
