In a combined repeated dose toxicity study with the reproduction/developmental toxicity screening test by oral administration to rats (OECD TG422, GLP), lower values in albumin and total protein in females in the groups of 200 and 1,000 mg/kg, and higher values in ALT and lower values in blood glucose in males in the 1,000 mg/kg group were observed, and as for organ weights, higher values in relative liver weights in males in the groups of 200 and 1,000 mg/kg, and higher values in relative kidney weights in males in the 1,000 mg/kg group were found. Histopathological examination revealed centrilobular hepatocellular hypertrophy of the liver in males in the groups of 200 and 1,000 mg/kg, tubular dilatation and regeneration epithelium in the kidney in males and females in the 1,000 mg/kg group, and increased hyaline droplets in the proximal tubules in males in the 1,000 mg/kg group (Safety Test (Ministry of Economy, Trade and Industry (METI, 2009))). From the above, toxicity was seen in the liver and kidney as a result of repeated administration, and kidney toxicity was at 1,000 mg/kg/day (converted guidance value: 467 mg/kg) above the guidance value range. However, because liver toxicity was observed at or above 200 mg/kg/day (converted guidance value: 93 mg/kg) corresponding to the guidance values for Category 2, it was classified in Category 2 (liver).
