However, the DFG's classification was made by analogy with the analogous compound (thiourea) and the evidence for this substance itself was inadequate in both epidemiological studies and animal experiments. [Reference Data, etc.] (1) As for the classification results by international organizations, the DFG classified it in Category 3 (List of MAK and BAT values 2020 (Accessed Oct. 2021)). In addition, the IARC classified it in Group 3 (IARC (1984)), the ACGIH classified it in A4 (ACGIH (2001)), and the ECHA classified it in Carc. 2 (EU-CLP Classification Results (Accessed Oct. 2021)). (2) The DFG classified this substance in Category 3 as is the case with the analogous compound thiourea (CAS RN 62-56-6) because thiourea causes thyroid cancer at high doses and has a similar structure and metabolism to this substance (DFG MAK (2011)). (3) In a carcinogenicity study for the analogous compound thiourea using rats, thyroid follicular cell adenoma/carcinoma, Zymbal gland tumor, and monocytic leukemia significantly increased, and in a carcinogenicity study with mice, liver cancer, lymphoma, thyroid follicular cell adenoma/carcinoma, hepatocellular adenoma/carcinoma, and adenoma/carcinoma of the adenohypophysis significantly increased (Initial Risk Assessment Report (NITE, CERI, NEDO, 2008)). (4) In a carcinogenicity study for this substance using rats and mice, no evidence for the carcinogenicity was obtained, but it was concluded that the study itself was inadequate to evaluate the carcinogenicity of this substance (IARC 30 (1982), ACGIH (7th, 2001), DFG MAK (2011)). (5) For the analogous compound thiourea, based on the data of (4) and since the IARC classified it in Group 3 because the evidence for the carcinogenicity in experimental animals was limited, it was classified in Category 2 in Japan (GHS classification result in FY2017). |
