In a 28-day repeated dose toxicity test by oral administration to rats (Guidelines specified by the Chemical Substances Control Law, GLP), decreases in hemoglobin level, erythrocyte count, and hematocrit value and increased methemoglobin were observed at or above 60 mg/kg (converted guidance value: 18.7 mg/kg/day) corresponding to the guidance value range for Category 2, histopathological examination revealed increased deposits of hemosiderin in the spleen and increased hematopoiesis in the bone marrow, and these were considered to be increased hematopoiesis to compensate for anemia with methemoglobinemia (JECDB (Access on July 2012)). Therefore, it was classified in Category 2 (blood). And at 300 mg/kg (converted guidance value: 93.3 mg/kg/day) corresponding to the guidance value range for Category 2, an increase in urinary protein, increases in erythrocytes and leukocytes and an increase in small round epithelial cells in urinary sediment, and an increase in urea nitrogen were found, and histopathological examination showed basophilic changes in the renal tubules and mineralization of the inner medulla, increased incidence of eosinophilic bodies in the renal tubular epithelium in addition to papillary necrosis, dilatation of the renal tubules, and cellular infiltration in the cortical interstitium (JECDB (Access on July 2012)). Therefore, it was classified in Category 2 (kidney). Furthermore, it was classified in Category 2 (liver) because increases in activities of gamma-GTP and GPT, an increase in total cholesterol, hypertrophy of centrilobular hepatocytes, and bile duct proliferation at the histopathological examination were seen at 300 mg/kg, and these were considered to be slight hepatic disorders (JECDB (Access on July 2012)).
