H302 | P301+P312 P264 P270 P330 P501 |
Based on the LD50 value of 743 mg/kg bw for rats (HSDB (2002)), the substance was classified as Category 4. | FY2009 | GHS Classification Guidance by the Japanese Government (March, 2009) |

Based on a LC50 value of 290 mg/m3/2h (0.205 mg/L/4h = 37 ppm) for mice (HSDB (2002)), the substance was classified into Category 1. Since the LC50 value (0.29 mg/L/2h) was lower than 90% of saturated vapour pressure concentration (0.49 mg/L), the classification criteria for gas was adopted. | FY2009 | GHS Classification Guidance by the Japanese Government (March, 2009) |

2 | Skin corrosion/irritation | Category 2 |
Warning |
H315 | P302+P352 P332+P313 P362+P364 P264 P280 P321 |
Based on a report of "severe skin irritant" as a human health effect (HSDB (2002)), the substance was classified into Category 2. As relevant information, the result in a standard Draize test with rabbits was "moderate" (RTECS (2006)). | FY2009 | GHS Classification Guidance by the Japanese Government (March, 2009) |
3 | Serious eye damage/eye irritation | Category 2 |
Warning |
H319 | P305+P351+P338 P337+P313 P264 P280 |
Based on a report of "severe eye irritant" for human (HSDB (2002)), the substance was classified into Category 2. As relevant information, there was a report that the result in a rabbit standard Draize test was "moderate" (RTECS (2006)). | FY2009 | GHS Classification Guidance by the Japanese Government (March, 2009) |

5 | Germ cell mutagenicity | Classification not possible |
- |
- | - | Classification not possible due to lack of data. From in vitro mutagenicity tests, there are reports of a negative Salmonella typhimurium strain TA100 mutation assay and a negative Chinese hamster V79 cell gene mutation test (IARC 27 (1982)). From in vivo tests, found as information in List 3, there is a report of a negative intraperitoneal injection mouse DNA damage test (in vivo genotoxicity test) (RTECS (2006)). In addition, the substance is an existing chemical substance, for which mutagenicity was established under the provisions of Article 57-3 of the Industrial Safety and Health Law. | FY2009 | GHS Classification Guidance by the Japanese Government (March, 2009) |
6 | Carcinogenicity | Not classified |
- |
- | - | Based on the classification of "Group 3" in IARC (IARC suppl. 7 (1987)), the substance was classified as "Not classified". As relevant information, in a test where male and female mice and male rats were fed diets containing 2,4,6-trimethylaniline hydrochloride for 15 months and observed for an additional 3 or 6 months, significant increases in the incidences of hepatomas in male and female mice and significant increases in the incidences of liver, lung and stomach tumours in male rats were observed. However, the Working Group noted that poor survival and the scant detail in the reporting make evaluation of the test difficult. (IARC 27 (1982)). | FY2009 | GHS Classification Guidance by the Japanese Government (March, 2009) |

Workers who had been engaged for eleven to twenty years in manufacture of the substance and aniline were reported to have narrowed peripheral vision fields, increase in size of blind spot and decrease in photosensitivity, for which the substance was thought to be responsible (HSDB (2002)). Additionally, there is a report that proved particular toxicity to retina when given to rats and rabbits, which produced morphologic changes in retina (HSDB (2002)). Based on a report of adverse effects for the eye in humans as information found in a document in List 2, the substance was classified into Category 2 (eye). | FY2009 | GHS Classification Guidance by the Japanese Government (March, 2009) |
