- - In the acute oral toxicity study in rats (OECD TG423, GLP-compliant), no treatment-related abnormalities were found in mortality, clinical signs of toxicity and necropsy at 300 mg/kg. However, emaciation, lateral position, hunchback position, decreased locomotor activity, bradypnea was observed at 2000 mg/kg, and all animals in dead or sacrificed at moribund stage revealed hemorrhage of glandular stomach, and protrusion area and thickening of the wall of the forestomach at necropsy. Even in the survivors, the protrusion area was also found ("Toxicity Testing Reports of Environmental Chemicals" (Chemicals Investigation Promoting Council) (2004)), however, it was described that these changes were considered as a cause of death or exhaustion probably attributed by treatment-related irritation to gastric mucosa ("Toxicity Testing Reports of Environmental Chemicals" (Chemicals Investigation Promoting Council) (2004)). Furthermore, the small size of the thymus and spleen in the decedents, as well as the small size of the thymus in the survivors, were observed, but it was also reported that these changes were considered as the effects attributed to exhaustion or stress ("Toxicity Testing Reports of Environmental Chemicals" (Chemicals Investigation Promoting Council) (2004)). Based on the information, because the effects on the stomach were regarded as the local effects and the cause of deaths were attributed to stress, these findings were not adopted as the basis to classify. In the combined repeated dose toxicity study with the reproduction/developmental toxicity screening test by oral administration in rats (OECD TG 422, GLP-compliant), increase in the spleen weight was observed in both sexes of 30 mg/kg/day group ( converted value as that of 90-day study: approx. 13 mg/kg/day), accompanied histopathologically by hemosiderin deposition and enhancement of erythroid extramedullary hematopoiesis in the spleen in both sexes, and enhancement of erythroid hematopoiesis in the bone marrow of the femur in females. It was reported that hematological changes considered as related to hematopoietic system involved those suggesting anemia such as low values of the erythrocyte count in both sexes, low value of hemoglobin concentration and high value of reticulocyte count in males ("Toxicity Testing Reports of Environmental Chemicals" (Chemicals Investigation Promoting Council) (2004)).
