In 2-generation mixed diet administration tests using rats, decreased number of births and pregnancies, and an increased number of stillbirths were observed in F0 and F1 generations in the highest dose groups; and atrophy of the prostate, decreased spermatozoa, necrotic spermatocytes of the epididymides, and atrophy of the testes were detected in male rats in the highest dose groups. In addition, oral administration tests were conducted during the organogenic period using either pregnant rats or pregnant rabbits. In either test, signs of maternal toxicity were observed in low dose groups, and an increased resorption, decreased fetal weight and increased rate of fetal mortality were also observed while no signs of teratogenicity were found (JMPR (1992)). In 3-month mixed diet tests using dogs, an increased liver weight, centrilobular or midzonal hepatocellular hypertrophy, enlargement of periportal hepatocytes, and chronic nephritis were observed at 1600 ppm (58.8 mg/kg/day), which falls under Category 2 guidance values (JMPR (1992)). Based on this report, the substance was classified into Category 2 (kidney). Additionally, in 3-month mixed diet administration tests, hepatic centrilobular hypertrophy and necrosis were detected at >= 1000 ppm (132 mg/kg/day for male and 232 mg/kg/day for female) in mice and at >= 3000 ppm (158 mg/kg/day for male and 195 mg/kg/day for female) in rats; in both studies, doses administered were above Category 2 guidance values (JMPR (1992)).
