From an LD50 value of 600 mg/kg for mice (RTECS (1997): the original literature, Pharmazie. 30, 147, 1975), it was classified in Category 4. | FY2011 | GHS Classification Guidance by the Japanese Government (July, 2010) |

3 | Serious eye damage/eye irritation | Classification not possible |
- |
- | - | No data. Besides, the information on 4-ethylphenol was reported. Severe ocular irritation that consists of moderate to severe conjunctival irritation (redness, chemosis, discharge, necrosis), iritis, corneal opacity, stippling, and ulceration was shown 24 hours after application of 0.1 mL/kg to four rabbits. The severe irritation was still evident in all animals until seven days after application. And one animal exhibited pannus and one animal exhibited hair loss around the eye. (HSDB (2003)) |

- | - | No data. Besides, a combined repeated dose toxicity test with reproductive/developmental toxicity screening test (OECD TG 422, GLP) in which a mixture of 3 isomers including this substance was orally administered to rats reported that general toxicity such as decreases in food consumption and weight gain in males and salivation in females was observed. But no adverse effects of test substance administration on sexual function and fertility were observed (US EPA HPV (2006)). |

8 | Specific target organ toxicity - Single exposure | Classification not possible |
- |
- | - | No data. Besides, a test in which rats were orally dosed a mixture of 3 ethylphenol isomers including 25.9% of this substance (OECD TG 425, GLP) reported that signs such as hypersalivation, blepharoptosis, decreased spontaneous motility, ataxia, tachypnea, righting reflex, anomalies in limbs observed at 1750 mg/kg immediately after dosing disappeared by seven days after administration. (USEPA/HPV (2006)) In addition, an oral dose of 2000 mg/kg of 4-ethylphenol in rats reported that no death in both males and females was observed and observed signs of staggering gait followed by proneness or recumbency, and tachypnea disappeared by 2 hours after dosing. After that, no symptoms by day 14 and no dose-related abnormalities in both males and females in necropsy were observed. (Report on Toxicity Tests of Chemical Substances, MHLW (Access on Aug. 2010)) |

9 | Specific target organ toxicity - Repeated exposure | Classification not possible |
- |
- | - | No data. Besides, in a combined repeated dose toxicity test with reproductive/developmental toxicity screening test (OECD TG 422, GLP) in which rats were orally administered a mixture of 3 isomers at 30 to 245 mg/kg/day including this substance, no dose-related effects except dirty hair, decreases in food consumption and a weight gain in males, and salivation in females were reported. (US EPA HPV (2006)) In addition, in a 28-day repeated oral administration test of 4-ethylphenol to rats, increased liver weight, squamous cell hyperplasia of a forestomach in pathological histology at 300mg/kg or higher, and additionally, staggering gait, symptoms like salivation, low body weight and food consumption, and increased kidney weight at 1000 mg/kg were observed. All changes recovered or showed recovery tendencies at the end of a recovery period. NOELs under conditions of this test were reported 100 mg/kg/day for males (converted to a 90-day equivalent: 31.1 mg/kg/day) and 300 mg/kg/day for females (converted to a 90-day equivalent: 93.3 mg/kg/day).
