- - Many epidemiological studies in workers in inhalation exposure to dust of this substance were reported.
It is written that workers exposed for over five years to the environment where levels measured as total dust (particles of 1 to 6 micro m constitute 4.5 to 30%) exceeded 10 mg/m3 more than 60% time to measure a work environment concentration were diagnosed of benign pneumoconiosis in X-ray examination, and mild pneumoconiosis (diagnosed by X-ray examination), respiratory symptoms, and slightly decreased pulmonary function were observed in workers exposed to particles of this substance as respirable dust at a 10 (mg/m3 x-years) concentration (ACGIH (7th, 2008)). However, there is no report clearly showing an increase of respiratory system tumors in exposure to this substance.
Also in experimental animals, inhalation exposure (six hours/day, five days/week) to this substance (90% of particles were not larger than 1.5 micro m in diameter) in rats, guinea pigs, or monkeys at a 13 mg/m3 concentration for 22 months resulted in no lung tumor formation in any species. What found was the only existence of particles of this substance in alveolar macrophages (ACGIH (7th, 2008)).
From the above, ACGIH classified in A4. Besides, IARC classified in Group 3 (IARC vol. 19, Suppl. 7 (1987)).
Therefore, also in this evaluation, the substance was classified as "Classification not possible" in this hazard class.

- - It is reported in ACGIH (7th, 2008) that Finnish female workers in occupational exposure to this substance did not show increased risk of spontaneous abortions, and that an increased odds ratio (2.3, 95% confidence interval: 1.0 to 5.1) for stillbirths or cases having infants with deaths after birth, malformations, or low birth weight was observed in a case-control study in Norwegian and Swedish female workers who were engaged in work handling this substance. However, other than these, there is neither report on reproductive effects in human by exposure to this substance nor reproductive/developmental toxicity information in experimental animals (ACGIH (7th, 2008), DFGOT vol. 2 (1991), PATTY (6th, 2012), HSDB (Access on June 2015)). Therefore, the classification is not possible due to lack of data.

As for humans, it is written that in epidemiological studies in workers in inhalation exposure to dust of this substance, workers exposed for over five years to the environment where levels measured as total dust (particles of 1 to 6 micro m constitute 4.5 to 30%) exceeded 10 mg/m3 more than 60% time to measure a work environment concentration were diagnosed of benign pneumoconiosis in X-ray examination, and mild pneumoconiosis (diagnosed by X-ray examination), respiratory symptoms, and slightly decreased pulmonary function were observed in workers exposed to particles of this substance as respirable dust at a 10 (mg/m3 x-years) concentration as cumulative exposure (ACGIH (7th, 2008)).
As for experimental animals, in a 22-month inhalation exposure test (an average diameter: 1.2 micro m) using rats, guinea pigs, or monkeys, phagocytosis of PVC by macrophages and benign pneumoconiosis (fibrosis, inflammation, deficits in pulmonary function, no cancer observed) were reported at 13 mg/m3 (0.013 mg/L). In a 15-week to 8-month inhalation toxicity test using rats (an average diameter: 1.31 micro m, 1.7 micro m), phagocytosis of PVC by macrophages, alveolar septal thickening, interstitial inflammation including the proliferation of connective tissue, and granulomatous foci characterized by the small nodular formation in the lung lymph nodes were reported at 8 to 12 mg/m3 (0.008 to 0.012 mg/L). On an oral route, it is reported that significant effects were not observed in a 2-year repeated oral administration toxicity test using rats did. Besides, it is reported that hyperplasia of the gastric mucosa was observed in a drinking water administration toxicity test using rats after ten months, but a period and amount of administration and so on were not described (ACGIH (7th, 2008)). Findings in the lung in animal tests were within the range of Category 1.
As above, as effects of inhalation exposure to dust of this substance, there is the report of benign pneumoconiosis, respiratory symptoms, and slightly decreased pulmonary function in humans.
On top of that, because effects on the lung were reported within the range of Category 1 in experimental animals using respirable-sized dust, ACGIH (7th, 2008) recommended a threshold limit value (TLV) of 1 mg/m3 for the respirable-sized fraction. Therefore, the substance was classified in Category 1 (respiratory organs).
