As for in vivo, among tests with mouse bone marrow cells, it was positive in a micronucleus test, equivocal in a chromosomal aberration test, and positive in a sister chromatid exchange test (NTP DB (Access on September 2013)). As for in vitro, it was positive in a bacterial reverse mutation test, a chromosomal aberration test and a mouse lymphoma test with cultured mammalian cells (NTP DB (Access on September 2013)). This substance was classified in Category 1A because it is classified in Group 1 by IARC (IARC 100F (2012)), A1 by ACGIH (ACGIH (1987)), K by NTP (NTP RoC (1980)), and Carc. 1A in EU CLP (ESIS (Access on Sept 2013)). Besides, urinary tract tumors due to work involving exposure to beta-naphthylamine, a carcinogen, are included in Disease designated by the Minister of Health, Labour and Welfare under the Industrial Safety and Health Act in Japan, and its manufacture is prohibited. It is concluded in IARC 100F (2012) that there was sufficient evidence in humans for the carcinogenicity of this substance in the urinary bladder. It is reported that many cases of bladder cancer in workers exposed to this substance were found, and the cumulative incidence was 25%. Furthermore, as for the relationship between exposure of workers engaged in manufacturing and use of this substance and the risk of urinary bladder cancer, 11 cohort studies in the United States, the United Kingdom, Japan, Poland, Russia, and Italy are available, all of which showed that the risk of human bladder cancer was significantly increased. For example, 26 bladder-cancer deaths were observed in British dyestuff-industry workers exposed to this substance, with an expected value of 0.3 (Standardized Mortality Ratio (SMR) 86.7, 95% CI: 56.6-127.0), and 58 cases of bladder cancer were found in the British rubber industry workers from 1946 to 1949 when antioxidants containing this substance were used (Standardized Incident Ratio (SIR) 1.71; 95% CI: 1.3-2.21).
