It was reported to be positive in a micronucleus test with bone marrow after oral administration to mice (in vivo somatic cell mutagenicity test) (RTECS (2003): the original article, Mutation Research: 208, 61, 1988), and weak responses were observed at multiple sampling time points, although it is a finding with one dose of 40 mg/kg. Therefore, it was classified in Category 2. Besides, as for in vitro tests, it is reported that it was negative in an Ames test (JECDB (2004)), and positive in a chromosomal aberration test with CHL cells of Chinese hamsters (JECDB (2004)). - - In a combined repeated dose toxicity study with the reproduction/developmental toxicity screening test with rats (OECD TG422, GLP), changes in urine such as turbidity of urine and a decrease in urine specific gravity were observed at the observation of general conditions, but no abnormalities from treatment with this substance were observed in delivery and lactation condition in addition to mating results such as estrus cycle, copulation index, fertility index, and the average number of estrus until copulation, and all pregnant animals delivered live newborns in the treated group like the control group. And the administration of this substance did not affect developmental parameters such as the number of corpus lutea, the number of implantations, implantation index, the number of newborns, delivery index, the number of live pups, live birth index, and viability index, and no abnormal morphology was seen in any newborn (JECDB (2004)). Therefore, there was no evidence of adverse effects on sexual function/fertility, but data are insufficient on effects on the development of offspring including teratogenicity.
