The nephrotoxicity of this substance (2-BP) was investigated because it was a metabolite of bromobenzene, a known nephrotoxin (CICAD 66 (2005)). Following of an oral administration to rats at 750 mg/kg or 1125 mg/kg (equivalent to Category 2 of the guidance values), significant elevations in both protein concentration and numbers of the epithelial cells in urine, and reduction in renal glutathione concentration were found (CICAD 66 (2005)). In the repeated dose oral study (7 - 28 days, 30 or 150 mg/kg bw) with rats conducted concurrently with a single exposure study, similar but less remarkable effects to those seen in a single exposure (significant elevations in protein concentration and numbers of the epithelial cells in urine, reduction in renal glutathione concentration) were also observed. However, the transition from single to repeated exposure did not result in exacerbation of nephrotoxicity (CICAD 66 (2005)).
