1 | Acute toxicity (Oral) | Classification not possible |
- |
- | - | Classification not possible due to lack of data. An LD50 value of 1050 mg/kg for rats is reported (GESTIS (Access on August 2016)), however, the data is from a List 3 information source, and details of the data are not available because the original report was company-owned data. Therefore, the information was not adopted for evidence of classification. Besides, for the following health hazards, please also see the structural isomers (2,3-dichlorotoluene (CAS RN 32768-54-0) and 2,4-dichlorotoluene (CAS RN 95-73-8)). |

- |
- | - | Based on the test results as non-irritant after applying 0.5 mL of the test substance for 4 hours on rabbit skin (OECD TG 404) (ECHA (Access on December 2016)), it is classified as "Not classified." | FY2016 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |
3 | Serious eye damage/eye irritation | Classification not possible |
- |
- | - | Classification not possible due to lack of data. Besides, it is described that in a standardized eye irritation test using rabbits, the mean irritating value was 4.4 for the conjunctiva, cornea and iris 24 hours to 8 days after application, and this substance was assessed to be non-irritating (GESTIS (Access on August 2016)). However, it was classified as "Classification not possible" because there is no other information. |

7 | Reproductive toxicity | Category 2 |
Warning |
H361 | P308+P313 P201 P202 P280 P405 P501 |
In a combined repeated dose toxicity study with reproduction/developmental toxicity screening test using rats orally administered, general toxicological findings, such as tubule vacuolation at 100 mg/kg or more in females, found dead, suppressed body weight gain, decreased food consumption, and histopathological changes in the kidneys (tubule vacuolation and regeneration) at 300 mg/kg in males were observed. However, fertility of parent animals was not affected at up to the highest dose (300 mg/kg). Meanwhile, lowered values for the number of live births, tendencies for a decreased birth index, tendencies for a decreased number of live pups on day 4 at 100mg/kg or more, and lowered body weights (only at 300 mg/kg) in pups were observed (Safety Test (Ministry of Economy, Trade and Industry (METI)) (Access on July 2016)). Since adverse effects in the live pups were observed at the dose level which showed maternal toxicity (renal toxicity at 100 mg/kg/day), the substance was classified in Category 2. | FY2016 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |
8 | Specific target organ toxicity - Single exposure | Classification not possible |
- |
- | - | Classification not possible due to lack of data. There is no available data on single dose exposure study of this substance in humans. In experimental animals, sedation, narcosis, reduced general health and loss of body weight were reported after single oral administration to rats (GESTIS (Access on August 2016)). However, the substance was classified as "Classification not possible" because the data is from a List 3 information source, and the original report was not available so that the details of the data are not available. |

9 | Specific target organ toxicity - Repeated exposure | Category 2 (kidney) |
Warning |
H373 | P260 P314 P501 |
There were no available data for humans. As for experimental animals, in a combined repeated dose toxicity test with reproductive/developmental toxicity screening test using rats administered by gavage (OECD TG 422), renal tubule vacuolation at 100 mg/kg/day equivalent to Category 2 (converted guidance value: 46.7 mg/kg/day) and regenerative renal tubules and renal tubular dilatation at 300 mg/kg/day equivalent to more than Category 2 (converted guidance value: 140 mg/kg/day) were observed. In the recovery group, lymphocytic infiltration and necrosis were found (Safety Test (Ministry of Economy, Trade and Industry (METI)) (Access on July 2016)). Therefore, the substance was classified in Category 2 (kidney). |

From 48-hour ErC50 = 1.4 mg/L for algae (Pseudokirchneriella subcapitata) and 48-hour EC50 = 1.4 mg/L for crustacea (Daphnia magna) (both Results of Aquatic Toxicity Tests of Chemicals conducted by Ministry of the Environment in Japan (Ministry of the Environment, 2009)), it was classified in Category 2. Besides, as for the test on algae, the 48-hour toxicity value conducted taking account of closed system test was used. | FY2016 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |
11 | Hazardous to the aquatic environment Long term (Chronic) | Category 2 |
- |
H411 | P273 P391 P501 |
If chronic toxicity data are used, then it is classified in Category 2 due to not rapidly degradable (Non-biodegradable, a degradation rate by BOD: 0 % (Biodegradation and Bioconcentration Results of Existing Chemical Substances under the Chemical Substances Control Law, 2008)), and 48-hour NOEC = 0.23 mg/L for algae (Pseudokirchneriella subcapitata) (Results of Aquatic Toxicity Tests of Chemicals conducted by Ministry of the Environment in Japan (Ministry of the Environment, 2009)). Besides, as for the test on algae, the 48-hour toxicity value conducted taking account of closed system test was used. If acute toxicity data are used for a trophic level for which chronic toxicity data are not obtained, then it is classified in Category 2 due to being not rapidly degradable (Non-biodegradable,a degradation rate by BOD: 0 % (Biodegradation and Bioconcentration Results of Existing Chemical Substances under the Chemical Substances Control Law, 2008)), and 48-hour EC50 = 1.4 mg/L for crustacea (Daphnia magna) (Results of Aquatic Toxicity Tests of Chemicals conducted by Ministry of the Environment in Japan (Ministry of the Environment, 2009)). It was classified in Category 2 from the above results. |
