When this substance was administered by gavage to female rats from 14 days prior to mating to day 3 of lactation, in maternal animals, as toxic symptoms, urine stains, a decrease in locomotor activity, pale skin, rough fur, etc. at or above 225 mg/kg/day, and unsteady gait at 750 mg/kg/day were observed. As for effects on the pups, a decrease in the live birth index and decreased number of alive pups by day 4 of lactation were observed (ACGIH (7th, 2009), Environmental Risk Assessment for Chemical Substances Vol.7, Tentative Hazard Assessment Sheet (Ministry of the Environment, 2009)). With regard to humans, it is described that this substance was used as an anesthetic in the 19th century (ACGIH (7th, 2001)). As for experimental animals, central nervous system depression was reported to be a toxic symptom in a single oral dose test with rats (ACGIH (7th, 2009), PATTY (6th, 2012)). As for the inhalation route, it was reported that in a test with mice to evaluate the irritation potential of this substance, when a decline in the respiration rate following short-duration (5 minutes) inhalational exposure to the vapor was measured, an RD50 value was 500 mg/m3 (ACGIH (7th, 2009), PATTY (6th, 2012)). From the above, it was classified in Category 3 (respiratory tract irritation, narcotic effects). Besides, it was reported that in a test with mice, administration by subcutaneous or intravenous injection showed hypnotic effects, and in intraperitoneal administration, clear hypnotic effects were observed at 400-500 mg/kg (ACGIH (7th, 2009)). Also, it is reported that a 4-week repeated toxicity test with rats dosed by gavage, hyaline droplet nephrosis in the males at or above 75 mg/kg/day (converted guidance value: 23 mg/kg/day) within the guidance value range for Category 2, urine stains and salivation at or above 225 mg/kg/day (converted guidance value: 70 mg/kg/day), and a low value of body weights, reduced forelimb grip strength, a decrease in locomotor activity, and unsteady gait at 750 mg/kg/day (converted guidance value: 233 mg/kg/day) exceeding the guidance value range for Category 2 were reported (Environmental Risk Assessment for Chemical Substances Vol.7, Tentative Hazard Assessment Sheet (Ministry of the Environment, 2009), ACGIH (7th, 2009)). Other than these, it was reported that in a 14-day inhalation toxicity study with mice exposed to the vapor, no effects on the trachea were observed at 300 ppm (converted guidance value: 0.23 mg/L) within the guidance value range for Category 2 (PATTY (6th, 2012)).
From the above, effects on the nervous system were reported, but these were observed at a dose exceeding the guidance value range for Category 2.
Therefore, it was classified as "Classification not possible."
Besides, since new information sources were used, the classification result was changed from the previous classification.
