(1) In a two-generation reproduction study with rats dosed by feeding, at a dose at which no parental toxicity was observed, a decrease in implantation rate was observed in F1 females, and at a dose (800 ppm) at which parental toxicity (reduced body weight gain, stricture of the liver due to intimal thickening of the intrahepatic portal branch) was observed, a decrease in implantation rate in P females, a decrease in the number of corpora lutea, and a decrease in the number of implantations in F1 females, a decrease in the number of viable fetuses and an increase in fetal mortality in F2 fetuses, reduced body weight gain and a delay in growth differentiation in F1 and F2 pups, and lower survival rate in F1 pups were observed (Risk Assessment Report (Pesticides) (Food Safety Commission of Japan, 2014)).
(2) In a developmental toxicity study with female rats dosed by gavage on days 6 to 15 of gestation, at a dose (300 mg/kg/day) at which a decrease in locomotor activity, involuntary movement of the head, ataxic gait, salivation, vaginal secretions, and reduced body weight gain were observed in dams, lower body weight, external malformations (encephalocele, rupture of the thoracic wall, rupture of the abdominal wall) and delayed ossification were observed in fetuses (Risk Assessment Report (Pesticides) (Food Safety Commission of Japan, 2014)).

[Evidence Data]
(1) In an acute oral toxicity test with rats (there was no description of the minimum dose at which effects were observed. It was assumed that effects were observed at least at doses lower than 1,000 mg/kg (within the range for Category 2) at which death began to occur), a decrease in locomotor activity, ataxia, a decrease in respiration rate, blunted response to contact and pain, hypodynamia, prone or lateral position, lacrimation, and dyspnea were observed (Risk Assessment Report (Pesticides) (Food Safety Commission of Japan, 2014)).
(2) In an acute dermal application test with rats (there was no description of the minimum dose at which effects were observed. It was assumed that effects were observed at least at doses lower than 1,600 mg/kg (within the range for Category 2) at which death began to occur), a decrease in locomotor activity, ataxia, a decrease in respiration rate, blunted response to contact and pain, hypodynamia, prone or lateral position, lacrimation and dyspnea were observed (Risk Assessment Report (Pesticides) (Food Safety Commission of Japan, 2014)).
(3) In a 4-hour inhalation exposure test with rats (powder, systemic exposure), at or above 1.11 mg/L, a decrease in locomotor activity, eyelid closure, salivation, and irregular breathing were observed (Risk Assessment Report (Pesticides) (Food Safety Commission of Japan, 2014)).

There was no report on hazard by repeated exposure to this substance in humans. Based on (1) to (4), effects on the nervous system at doses for Category 1, and blood system and liver at doses for Category 2 were observed in experimental animals. Therefore, it was classified in Category 1 (nervous system), and Category 2 (blood system, liver). With the addition of new information, the classification results were changed from the previous classification.

[Evidence Data]
(1) It was reported that, in a 90-day feeding test with rats, at or above 300 ppm (males/females: 21.2/22.7 mg/kg/day, both within the range for Category 2), an increase in reticulocyte count was observed in females; at or above 900 ppm (males/females: 63.1/67.7 mg/kg/day, both within the range for Category 2), unkempt fur and a decrease in ALP were observed, and furthermore, intimal thickening of the intrahepatic portal branch, hemorrhage in the medullary cord, and mastocyte infiltration of the mesenteric lymph nodes were observed in males, and an increase in white blood cell count (WBC) and increasesd in absolute and relative liver weight were observed in females; and at 1,200 ppm (males/females: 85.1/87.8 mg/kg/day, both within the range for Category 2), decreases in hemoglobin (Hb) and hematocrit (Ht) and hyperplasia of plasma cells in the medullary cord of the mesenteric lymph nodes were observed, and furthermore, a decrease in red blood cell count (RBC), an increase in reticulocyte count, an increase in ALT, and an increase in extramedullary hematopoiesis in the spleen in males were observed, and increases in platelet count (PLT) and lymphocyte count, intimal thickening of the intrahepatic portal branch, hemorrhage in the medullary cord, and mastocyte infiltration of the mesenteric lymph nodes were observed in females (Risk Assessment Report (Pesticides) (Food Safety Commission of Japan, 2014)).
(2) In a one-year oral toxicity test with dogs dosed by capsules, at 6 mg/kg (within the range for Category 1), tremor, tonic convulsions, salivation, lying on side, quickening of respiration, and gait difficulty were observed, and staggering gait in males and clonic convulsions in females were observed (Risk Assessment Report (Pesticides) (Food Safety Commission of Japan, 2014)).
(3) It was reported that, in a two-year feeding test with rats, at or above 600 ppm (males/females: 37.3/39.9 mg/kg/day, both within the range for Category 2), intimal thickening of the intrahepatic portal branch, liver atrophy, stomach edema and pigmentation in the small intestine were observed, and furthermore, regenerative renal epithelium was observed in females; and at 1,200 ppm (males/females: 83.7/88.2 mg/kg/day, both within the range for Category 2), decreases in Ht and Hb, necrosis of the liver, and atrophy of the pancreatic exocrine gland were observed, and furthermore, an increase in mortality, a decrease in RBC, and an increase in PLT were observed in males, and fibrosis of the heart was observed in females. Additionally, in a group of rats sacrificed in week 52 of this test, at or above 600 ppm (males/females: 37.3/39.9 mg/kg/day, both within the range for Category 2), intimal thickening of the intrahepatic portal branch was observed, and furthermore, stomach edema and pigmentation in the small intestine were observed in females; and at 1,200 ppm (males/females: 83.7/88.2 mg/kg/day, both within the range for Category 2), a decrease in Ht and liver atrophy were observed, and furthermore, a decrease in RBC, an increase in neutrophil percentage, a decrease in lymphocyte percentage, and pigmentation in the small intestine were observed in males, and a decrease in Hb and increases in PLT and WBC were observed in females (Risk Assessment Report (Pesticides) (Food Safety Commission of Japan, 2014)).
(4) In a 28-day inhalation (dust) toxicity test with rats, at or above 120 mg/m3 (converted value in 90 days, 6 hours/day, 7 days/week: 0.032 mg/L, within the range for Category 2), an increase in nasal discharge and an increase in ALT were observed, and furthermore, a decrease in Ht in males and salivation, an increase in WBC, an increase in AST, and a decrease in ALP in females were observed; and at 360 mg/m3 (0.36 mg/L, exceeding Category 2), death (males: 9 cases, females: all cases), a decrease in locomotor activity, salivation, rough hair coat, myocarditis, thymic atrophy, spleen extramedullary hematopoiesis, hepatic congestion and centrilobular hepatocyte necrosis, granular matters in the renal tubules, pulmonary congestion, edema, and hemorrhage were observed, and in females, a decrease in muscle tone, piloerection, anemia, and urinary incontinence were observed (Risk Assessment Report (Pesticides) (Food Safety Commission of Japan, 2014), A pesticide abstract and evaluation report (Food and Agricultural Materials Inspection Center, 2014)).
