- - The classification was concluded as "Not classified" based on the negative results in the micronucleus test using bone marrow obtained from mice exposed by inhalation (in vivo mutagenicity test in somatic cells) (IUCLID (2000)). These results mentioned above were obtained from retrial study. It was described that ethers generally had narcotic action and it was indicated that structurally similar vinyl ethers (the substance and isopropyl vinyl ether) possessed depressive action of central nervous system and anesthetic action in the laboratory animals (HSDB (2002)). In addition, as symptoms following exposure to humans, headache, dizziness, excitation, loss of consciousness were described (HSDB (2002)). - - In the 28-day inhalation toxicity study in rats (OECD TG 412, GLP-compliant) exposed to the substance at the concentrations of 500, 3500 and 25000 ppm, slight prolongation of prothrombin time and slight decrease in total protein concentration were observed in males of all treatment groups. The signs consisting of restlessness and piloerection, and changes in the liver weight were observed in the highest dose level (25000 ppm), and suppression of body weight gain was observed in the doses of 3500 ppm or more. However, no remarkable changes were found at necropsy. The only histopathological change was atrophy of the nasal olfactory epithelium in the highest dose group. The NOAEL of 3500 ppm (converted concentration as that of 90-day study: 1089 ppm) was reported for females (IUCLID (2000)). For males, the effects at 3500 ppm were slight, and in another 28-day inhalation toxicity study in male rats (OECD TG 412, GLP-compliant), no effects as described above were noted, and the NOAEL of 1500 ppm (converted concentration as that of 90-day study: 467 ppm) was reported (IUCLID (2000)). Based on the information, since the substance was gasiform by definition of GHS and the NOAELs in the 28-day repeated inhalation studies in rats (OECD TG412, GLP-compliant) were over the upper limit (250 ppm) of the range of the guidance values, "Not classified" was justified.
