5 | Germ cell mutagenicity | Classification not possible |
- |
- | - | Because it was not possible to classify a substance as "Not classified" according to the revised GHS classification guidance for the Japanese government, it was classified as "Classification not possible." As for in vivo, a dominant lethal test in rats, a micronucleus test in mouse bone marrow cells, and an unscheduled DNA synthesis test in rat hepatocytes were negative (ECETOC JACC 48 (2005)). On the other hand, there is a positive result in a micronucleus test in mouse bone marrow cells (ECETOC JACC 48 (2005)). However, it is written in ECETOC JACC 48 (2005) that there is little concern of mutagenicity for this substance due to slight statistical significance in the frequency of micronucleus formation observed only in males at the highest dose of 1200 ppm. As for in vitro, there are data of a negative bacterial reverse mutation test (ACGIH (7th, 2010), ECETOC JACC 48 (2005)) and one positive gene mutation test (hprt) in cultured mammalian cells (ACGIH (7th, 2010)), but other 3 data were negative (ECETOC JACC 48 (2005)). A chromosomal aberration test in cultured mammalian cells was positive (ECETOC JACC 48 (2005)), but a positive result was observed only at the highest dose, and it was evaluated as weak clastogenic in ECETOC JACC 48 (2005). | FY2015 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |

8 | Specific target organ toxicity - Single exposure | Category 1 (central nervous system, respiratory organs, kidney) |
Danger |
H370 | P308+P311 P260 P264 P270 P321 P405 P501 |
There is no human information. As for experimental animals, this substance is irritating to the respiratory tract (ACGIH (7th, 2010)). In inhalation exposure in rats at 380-1,200 ppm, renal dysfunction and necrosis of proximal tubules were observed, but reversibility was reported. At these doses, there were no effects on other organs. In inhalation exposure in mice, guinea pigs, and rabbits at 2,000 ppm or higher, nephrosis was observed. In rabbits at 2,000-5,000 ppm, degeneration of kidney cells was reported (above ACGIH (7th, 2010), ECETOC JACC 48 (2005)). Pulmonary edema in rats in inhalation exposure at 500 ppm (LC50), central nervous signs such as loss of coordination and clonic convulsion, and necrosis of kidney cells in rats in inhalation exposure at 1,830 ppm (LC50) were reported (ECETOC JACC 48 (2005)). All of the above findings were observed at doses corresponding to Category 1. From the above, the substance was classified in Category 1 (central nervous system, respiratory system, kidney). The previous classification was revised by reviewing information. |
FY2015 | GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.1)) |
9 | Specific target organ toxicity - Repeated exposure | Category 1 (kidney) |
Danger |
H372 | P260 P264 P270 P314 P501 |
There is no human information. As for experimental animals, in a 2-week inhalation toxicity test using rats, mild nephropathy characterized by diffuse degeneration of the inner cortical tubules in the kidney was observed at 200 ppm (converted to a Guidance value equivalent: 20.3 ppm). Moreover, in a 90-day inhalation toxicity test using mice, regeneration of the inner cortical tubules, and cytomegaly and necrosis of the tubular epithelium in the kidney were found at 50 ppm (ECETOC JACC 48 (2005), ACGIH (7th, 2010)). Therefore, it was classified in Category 1 (kidney). |
