3 | Serious eye damage/eye irritation | Category 2B |
Warning |
H320 | P305+P351+P338 P337+P313 P264 |
"Slight irritation" was reported in eye irritation tests using rabbits with 10% suspension in corn oil and with powder (NTP TOX-28 (1993)). Therefore, it was classified into Category 2B. | FY2008 | GHS Classification Guidance by the Japanese Government (Sep, 2008) |
4 | Respiratory sensitization | Category 1 |
Danger |
H334 | P304+P340 P342+P311 P261 P284 P501 |
There is the following description in the List 1 (NTP TOX-28 (1993)). As human toxicities, several cases of the occupational disease (asthma) attributed to exposure to the substance have been reported in literature. In one case, five workers involved in the production of epoxy resins developed respiratory symptoms and physiologic abnormalities following the introduction of the substance into the product formation. Inhalation challenge tests with the substance in three of the five workers reproduced their symptoms and resulted in both immediate and delayed responses. Immunologic studies did not demonstrate specific antibodies; however, clinical signs suggested a hypersensitivity reaction rather than an irritant effect. The presence of IgE antibody specific to the substance was later reported in seven female workers exposed to the chemical via an epoxy resin; the antibody was identified following skin-prick and radioallergosorbent tests (radioactive allergen absorption test). Based on the above descriptions, the substance was classified into Category 1. | FY2008 | GHS Classification Guidance by the Japanese Government (Sep, 2008) |
4 | Skin sensitization | Classification not possible |
- |
- | - | No data available. | FY2008 | GHS Classification Guidance by the Japanese Government (Sep, 2008) |
5 | Germ cell mutagenicity | Not classified |
- |
- | - | In a chromosomal aberration test using mouse bone marrow (in vivo mutagenicity tests using somatic cells), the substance was negative (NTP TOX-28 (1993)), and therefore, it was classified into "Not classified". As relevant information, it is described that in sister chromatid exchange (SCE) tests (in vivo genotoxicity tests using somatic cells), the substance showed negative results, and the observed SCE frequencies increased dose-dependently, but they were small, and in one of the dose groups, the degree of increase in SCE frequencies was less than that of the control group (NTP TOX-28 (1993)). | FY2008 | GHS Classification Guidance by the Japanese Government (Sep, 2008) |
6 | Carcinogenicity | Classification not possible |
- |
- | - | No data available. | FY2008 | GHS Classification Guidance by the Japanese Government (Sep, 2008) |
7 | Reproductive toxicity | Classification not possible |
- |
- | - | It is described that in a developmental toxicity test in which rats were gavaged during the pregnancy period, including the organogenetic period, except for a slight increase in the incidence of skeletal malformation of fetuses in the 2000 mg/kg administration group, embryotoxicity and fetal toxicity, teratogenicity, and toxicities to parental animals were not detected (NTP TOX-28 (1993)). Also, it is documented that in an oral gavage test for 13-weeks using rats and mice, no abnormality of testis morphology or vaginal cells were observed (NTP TOX-28 (1993)). However, no data on reproductive toxicity is available, and due to the lack of data, classification is not possible. | FY2008 | GHS Classification Guidance by the Japanese Government (Sep, 2008) |
8 | Specific target organ toxicity - Single exposure | Classification not possible |
- |
- | - | No data available. | FY2008 | GHS Classification Guidance by the Japanese Government (Sep, 2008) |
9 | Specific target organ toxicity - Repeated exposure | Category 1 (lung) |
Danger |
H372 | P260 P264 P270 P314 P501 |
It is described that in an inhalation exposure test with dust or fumes of the substance for 13 weeks using rats, at the exposure range of 0.0005 - 0.05 mg/L/6h/day, hepatocellular hypertrophy and increased lung weights associated with multifocal accumulations of aveolar macrophages and alveolar hemorrhage were observed dose-dependently. Also, it is documented that in a test for four weeks, fumes at 0.04 mg/L/6h/day (90 day conversion, approximately 0.01 mg/L/6h/day) was recognized to adversely affected the lungs (NTP TOX-28 (1993)), and the dose is within the guidance values of Category 1, the substance was classified into Category 1 (lung). As relevant information, in an oral administration test by gavage for 13 weeks using rats, affects on the kidneys, for example, at the dose of 187 mg/kg/day, dilation of the renal tubular, at higher doses, necrosis were observed, and the NOAEL was 94 mg/kg/day. While it is described that in a test using mice, even at the dose of 1500 mg/kg/day, no histopathological change was observed (NTP TOX-28 (1993)). Since these doses are above the guidance values of Category 2, as for oral administration, the substance is classified into "Not classified". |
