In humans, there are reports of headache, vertigo and difficulty in breathing, and in experimental animals, narcosis, coordination ataxia and loss of reflexes were seen (ACGIH (7th, 2001), PATTY (6th, 2012)). In humans, there is no knowledge on repeated exposures. As for experimental animals, there are descriptions that in a combined repeated dose toxicity study with the reproduction/developmental toxicity screening test (OECD TG 422; GLP-compliant)in which rats were repeatedly exposed to the vapor of this substance for 49 days for males and for 36-49 days until day 4 of lactation for females, including 14 days before mating for males and females that exposure-dependent histopathological changes in the respiratory and olfactory epithelium of the nasal passages were observed (SIDS (2011)), and that irritating effects on the respiratory organs as histological change, and nasal exudate, irritant changes in the respiratory and olfactory epithelium, chronic inflammation or focal metaplasia of the epithelium as the inflammatory change were seen (PATTY (6th, 2012)). There is a report that the LOAEL was 31 ppm (124 mg/m3, converted guidance value: 0.068 mg/L/6 hours (Category 1)), where exudate in the olfactory epithelium was the only finding (SIDS (2011)). Decreased body weight gain was observed as systemic effects in this test, but no findings suggesting target organs other than the respiratory organs were seen (SIDS (2011), PATTY (6th, 2012)). There were no other reliable data available for classification.
Therefore, from the results of the inhalation exposure test with rats, it was classified in Category 1 (respiratory organs). Besides, the findings in humans in ACGIH (7th, 2001) were described as evidence of the classification in the previous classification, however, it was judged that it could not be used for this classification because it is a description of the effects of acute exposure.
