As relevant information, as for aluminum hydroxide, no maternal toxicity, embryo/fetal toxicity or teratogenicity were reported in the oral study in mice administered during the gestation period (66.5 - 266 mg/kg/day), and no toxic effects were reported in the oral study in mice administered during the gestation period (57.5 mg/kg/day). Reduced fetal body weight and increased skeletal variations were reported in the oral study in rats administered during the gestation period (384 mg/kg/day). It was also reported that in the oral study in rats administered during the gestation period (192 - 768 mg/kg), the NOEL was 768 mg/kg/day (the aforementioned, JECFA FAS 58 (2007)). As for the current classification for 2-buthanol was "In the two-generation reproduction study by drinking water in rats, there was no effect on reproductive potential, only the minimum influence (growth retardation of pups) was observed. On the other hand, in the inhalation study in rats exposed during the gestation period, reduction of live newborns and increase of embryo absorption was observed at the dose in which toxic effects on dams were observed (PATTY, 4th (1994), ACGIH (2002), DFGOT vol.19 (2003)). "Inhalation by high concentration exposure levels to mice caused the symptoms of depression of central nervous system (DFGOT vol.19 (2003)). By inhalation exposure, anesthetic actions were seen in humans and mice (EHC 65 (1987), and in rats and mice (PATTY, 4th (1994), DFGOT, vol.19 (2003)). From the description (ACGIH (2002), DFGOT, vol.19 (2003), Society for Occupational Health, Recommendation of Occupational Exposure (1986)) that nasal and throat were stimulated in humans in high exposure levels, the substance was considered to be respiratory irritant. Thus the substance was concluded as Category 3 (drowsiness and dizziness) and Category 3 (irritation to respiratory organs)." As relevant information on aluminum hydroxide, it was described that this was used as a pharmaceutical drug (antacid), and this might cause constipation, nausea, vomiting as its side effects (Handbook of Japanese Pharmaceutical Drugs (2010), Merck Manual, vol. 18 (2006)). It was also described that through a long-term treatment, this might cause aluminum encephalopathy or aluminum osteopathy (Handbook of Japanese Pharmaceutical Drugs (2010)). As for the animal experiments, no effects were reported on clinical signs, food and water consumption, growth, hematological and serum analyses, tissue and plasma concentrations of aluminum, histopathology in the 28-day oral dose study in rats (converted dose level as that of 90-day study: 407 mg/kg/day) (EHC, 194 (1997)). It was also reported that in the oral dose study in rats (17 - 29 mg/kg/day) the animals showed significant impairment of growth at 3 - 4 weeks of administration, and rachitic changes occurred in the bone (HSDB (2009)).
