In OECD TG403, nose-only exposure is recommended, and animal age is specified as 8-12 weeks. Therefore, by adopting > 5.15 mg/L (male), > 5.15 mg/L (female) from the test which is a later one and which test methods were appropriate, the substance was classified as "Not classified."
 In addition, the reference value of mist was applied because LC50 values are higher than the saturated vapor pressure concentration (0.0014 mg/L). On the basis of newly obtained information, the category was revised.

 Among 2-generation reproductive toxicity tests using rats by oral route (diet administration), in one test, abnormality in offspring was not observed at the dose (300 ppm) at which inhibition of red blood cell cholinesterase activity was found in F0, F1 parent animals, in the other test, at the dose (1,500 ppm) at which reduced weight gain, feed intake reduction, and increased relative liver weight, etc. were found in F0, F1 parent animals, reduced weight gain observed in F1, F2 pups, in addition delayed sexual maturation (delayed preputial separation, delayed vaginal opening) in F1 offspring were found (Risk Assessment Report (Pesticides) (Food Safety Commission, 2009)). In development toxicity tests using pregnant rats and pregnant rabbits by gavage administration (rats: 7 – 19 days of pregnancy, rabbits: 6 – 18 days of pregnancy), in rats, abnormality was not found in fetuses at the dose (100 mg/kg/day) at which maternal animal toxicity (salivation, increased absolute/relative liver weight) was observed, but in rabbits, increased post-implantation embryonic mortality was found at the dose (80 mg/kg/day) at which maternal animal toxicity (reduced weight gain, feed intake reduction) was observed (Risk Assessment Report (Pesticides) (Food Safety Commission, 2009)).
 As above, it was classified in Category 2 for this hazard class because in a 2-generation reproductive toxicity test in rats, growth/development suppression was observed in next generation at the dose at which toxic effects occurred in parent animals, and in the developmental toxicity test in rabbits, increased post-implantation embryonic mortality was observed at the dose at which maternal animal toxicity occurred. Besides, the classification was changed because it was based on new information source that was available after the previous classification.

 There is no data on single exposure in humans. In experimental animals, it is described that in a single oral dose study using rats and mice on this substance, bradykinesia, prone position, tachypnea, straub tail, convulsions, eyelid closure with lacrimation, and piloerection were observed but resolved at four days (rats) or three days (mice) after administration (A pesticide abstract and evaluation report (Food and Agricultural Materials Inspection Center, 2009), Risk Assessment Report (Pesticides) (Food Safety Commission, 2009)). Although minimal doses at which these symptoms appeared were not described, it is written that test doses were 360-1,548 mg/kg in rats and 592-4,827 mg/kg in mice, and a minimal lethal dose was 518 mg/kg in rats and 769 mg in mice (A pesticide abstract and evaluation report (Food and Agricultural Materials Inspection Center, 2009)). Therefore, it is thought that the doses where effects were seen are close to the minimal lethal dose which is within the range of Category 2. As for inhalation route, it is described that in a single 4-hour inhalation exposure study in rats, respiratory depression, sleep/coma, inanimation, piloerection, behavioral inhibition, back arch position, rough fur, crouching position, tremor, weakening, and gasping were observed, but survived animals recovered 13 days after exposure (A pesticide abstract and evaluation report (Food and Agricultural Materials Inspection Center, 2009), Risk Assessment Report (Pesticides) (Food Safety Commission, 2009)). Although minimal doses at which these symptoms appeared were not described, it is written that test doses were 0.25-1.48 mg/L and a minimal lethal dose was 0.80 mg/kg in males and 0.51 mg/kg in females (A pesticide abstract and evaluation report (Food and Agricultural Materials Inspection Center, 2009)). Therefore, it is thought that the doses where effects were seen are close to the minimal lethal dose which is within the range of Category 1. This substance is organophosphorus germicide, and organophosphorus pesticide (germicide) is generally known to cause effects on the nervous system in humans (IPCS, PIM G001 (1989)). From the above, it was classified in Category 1 (nervous system). The category was changed from the previous classification.
