Rationale for the classification Classification year (FY) GHS Classification Guidance for the Japanese Government
1 Acute toxicity (Oral) Not classified
-
-
- - Based on LD50 values of 7,120 mg/kg (males), 7,236 mg/kg (females), 7,954 mg/kg (males and females) (all SIDS (2010)), and 300-2,000 mg/kg (females) (SIDS (2010), JECDB (Access on July 2013)) for rats, it was classified as "Not classified" to which most data correpond. FY2013 GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.0))
1 Acute toxicity (Dermal) Not classified
-
-
- - It was classified as "Not classified" based on LD50 values for rabbits of about 3,880 mg/kg (males) and about 3,259 mg/kg (females) (SIDS (2010)). FY2013 GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.0))
1 Acute toxicity (Inhalation: Gases) Not classified (Not applicable)

It was classified in Category 4 based on a 4-hour LC50 value of 2,773 ppmV (male and female) for rats (SIDS (2010)). Besides, because LC50 was lower than 90% of the saturated vapour pressure concentration (20,263 ppmV), a reference value in the unit of ppmV was applied as vapour with little mist. FY2013 GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.0))
1 Acute toxicity (Inhalation: Dusts and mists) Classification not possible
-
-
- - Classification not possible due to lack of data. FY2013 GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.0))
2 Skin corrosion/irritation Classification not possible
-
-
- - Classification not possible due to lack of data. FY2013 GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.0))
3 Serious eye damage/eye irritation Not classified
-
-
- - It was classified as "Not classified" because it is described in SIDS (2010) that it was assessed to be not irritating in three tests in which this undiluted substance was applied to rabbits (OECD TG 405, etc.). FY2013 GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.0))

5 Germ cell mutagenicity Classification not possible
-
-
- - It was classified as "Classification not possible" because it was not possible to classify a substance as "Not classified" according to the revised GHS classification guidance for the Japanese government. As for in vivo, it was negative in a micronucleus test with mouse peripheral blood erythrocytes (SIDS (2010)). As for in vitro, it was negative in both a bacterial reverse mutation test (JECDB (Access on July 2013), SIDS (2010)) and a gene mutation test (hgprt gene locus) with cultured mammalian cells (SIDS (2010)), and positive in a chromosomal aberration test with cultured mammalian cells (JECDB (Access on July 2013), SIDS (2010)). From the above data, it is concluded in SIDS (2010) that this substance is not genotoxic. FY2013 GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.0))
6 Carcinogenicity Classification not possible
-
-
- - Classification not possible due to lack of data. FY2013 GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.0))
7 Reproductive toxicity Classification not possible
-
-
- - Classification not possible due to lack of data. Besides, in JECDB (Access on July 2013), data from a combined repeated dose toxicity study with the reproduction/developmental toxicity screening test were obtained, and only in the 1,000 mg/kg bw/day group, abnormalities in estrus cycles (lower number of estrous cases in 14 days before mating, 1,000 mg/kg bw/day group: 2.5 plus or minus 0.7, control group: 3.2 plus or minus 0.4) were observed, but there were no effects on copulation index and days needed for copulation, and no effects on fertility and offspring were seen. Furthermore, because 1,000 mg/kg bw/day was the dose where death in parent animals (2/12 animals in males, 1/12 animals in females) and lower body weights were observed, it was thought that the abnormalities in estrus cycles were slight changes related to general toxicity, not reproductive toxicity. However, the test is a screening test, and there was sufficient information on developmental toxicity, therefore it was classified as "Classification not possible.". Because in inhalation exposure in rats (vapour), hypoactivity, ataxia, prostration, tremors, and reflex disorders were observed at doses (11.9-32.5 mg/L) within the guidance value range (SIDS (2010)), it was classified in Category 2 (central nervous system). Furthermore, in oral administration to rats (970-31,040 mg/kg), reversible sluggishness, piloerection, unsteady gait, etc. were found (SIDS (2010)), and in dermal exposure in rabbits (1,940-7,760 mg/kg), reversible inanimation, unsteady gait, prostration were seen (SIDS (2010)). From the above results, it was classified in Category 3 (narcotic effects). FY2013 GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.0))
9 Specific target organ toxicity - Repeated exposure Category 2 (urinary bladder)

Warning
H373 P260
P314
P501
It is described that in a combined repeated dose toxicity study with the reproduction/developmental toxicity screening test with rats dosed orally, hyperplasia of the transitional epithelium in the urinary bladder was observed at the dose (62.5 mg/kg/day (converted guidance value: 29 mg/kg/day)) within a guidance value range for Category 2 (SIDS (2010), JECDB (Access on July 2013)). Therefore, it was classified in Category 2 (urinary bladder). Besides, in a test in which rats were exposed by inhalation to the vapour of this substance for 14 weeks, there were no findings to assign a category within a guidance value range, but effects on the kidney (papillary necrosis, interstitial edema, and papillary hyperplasia of the transitional epithelium) and urinary bladder (cystitis) were observed at the dose (2.4 mg/L/6 hours/day) corresponding to "Not classified" (SIDS (2010)). 10 Aspiration hazard Classification not possible

-
- - Classification not possible due to lack of data. FY2013 GHS Classification Guidance for the Japanese Government (FY2013 revised edition (Ver. 1.0))
