This substance was irritating to the respiratory tract (ACGIH (7th, 2001)). In addition, it induced pulmonary edema (ACGIH (7th, 2001), HSDB (Access on September 2014)). In humans, it was highly toxic by the inhalation route, and severe symptoms of pulmonary edema with chest pain, coughing, sputum, bluish discoloration of the skin, wheezing and difficulty in breathing were caused by exposure. Pulmonary edema had a delayed onset of several hours and then became rapidly worse, resulting in death (HSDB (Access on September 2014)). Also, in experimental animals, it acted like in humans and caused pulmonary edema. By a 4-hour exposure of rats, increases in serum AST and ALT activities, changes in conditioned reflexes (details unknown) and pulmonary edema at 0.24 ppm, dyspnea, hyperpnea and pulmonary hyperemia at 0.25 ppm were observed. And an increase in respiration rates was observed at 0.3 ppm (ACGIH (7th, 2001)). From the above, the target organ was determined to be the lungs, and in experimental animal tests, symptoms were observed at doses within the guidance value range of Category 1, therefore, it was classified in Category 1 (respiratory organs). |

In ACGIH (7th, 2001), in two tests in which rats were exposed by inhalation to this substance (the gas) at a concentration of 0.1 ppm for 10 days (6 hours/day, 5 days/week), while there is a description that exposure caused mild respiratory impairment and restlessness, and in some animals, cyanosis and moist rales were observed. No pathological abnormalities were observed at necropsy in any tests. Other than these, there were no known hazards by repeated exposure to this substance. However, this substance caused severe acute toxicity by the inhalation route (HSDB (Access on September 2014)). In addition, although the details were unknown, there is a description that this substance showed similar effects in animals and humans, and death cases occurred due to acute pulmonary effects including pulmonary edema (ACGIH (7th, 2001)), therefore, in repeated exposure, "respiratory organs" were considered to be the target organ. The introductory 10-day inhalation exposure test with rats corresponded to a 2-week exposure test. Considering that the minimum requirements for the administration duration (14 days (2 weeks) or more) were met as data available for classification, the converted value for the guidance value category was 0.015 ppm, and respiratory disorders were observed from the lower limit concentrations of Category 1, and it was well assumed that clear respiratory effects occurred at further higher concentrations within the range of Category 1 (<=50 ppm). Therefore, it was classified in Category 1 (respiratory organs). Besides, although it was classified in Category 1 (central nervous system) in the previous classification based on the results of a 19-week intermittent inhalation exposure test with rats listed in the information source in List 3 (RTECS), in this investigation, this test could not be found among information sources in List 1 (ACGIH), List 2 (HSDB), and List 3 (GESTIS, ICSC). |
