- - In a combined repeated dose toxicity study with the reproduction/developmental toxicity screening test by oral administration to rats (OECD TG 422, GLP, doses: 0, 12.5, 50, 200 mg/kg/day), in parent animals, reduced weight gain or a trend toward reduced weight gain was observed in males in the 200 mg/kg group during the latter part of administration period and in females during gestation and nursing period. On the other hand, as for reproductive function, no treatment effects were found in estrous cycle, the number of corpus luteum, the number of implantation sites, copulation index, fertility index, and duration of mating. And on examination on parturition and lactation, no effects of the test substance were seen in gestation length, gestation index, the number of newborns, live birth index, the number of live newborns, stillborn index, sex ratio, body weights of offspring on lactation days 0 and 4, and viability index on day 4, and examination on neonatal surface revealed no anomalies (JECDB (2001)). From the above results, no treatment-related adverse effects were shown on sexual function/fertility, but as for the development of offspring, there were no data on the examination of fetuses at the end of gestation. - - In a single administration toxicity test with rats (OECD TG 401, GLP), one male and two females died in the 2,000 mg/kg group, and on observation of general status, no abnormalities were seen on the day of the administration, but one day after the administration, bradypnea and lateral position were observed in one female in the 2,000 mg/kg group, and soiled perianal region and hypoactivity were found in males and females in the 2,000 mg/kg group. On pathological examination, no macroscopic abnormalities were seen in the organs in the surviving animals, one dead male in the 2,000 mg/kg group showed macroscopic and histological anomalies in the kidney and thymus, and one dead female in the 2,000 mg/kg group showed macroscopic and histologic changes in the lung (JECDB (2001)). From the above report, deaths of some animals were observed at 2,000 mg/kg, but the surviving animals did not have abnormalities, and it was hard to specify the target organ. In a combined repeated dose toxicity study with the reproduction/developmental toxicity screening test by oral administration to rats (OECD TG 422, GLP, doses: 0, 12.5, 50, 200 mg/kg/day), the incidence of hypertrophy of zona glomerulosa cells in the adrenal gland increased in males in the 200 mg/kg group (converted guidance value: 77.8 mg/kg/day) corresponding to the guidance value range for Category 2. From the description that hypertrophy of zona glomerulosa cells was considered to be caused by electrolyte imbalance, and an increase in urine volume also observed in males in the group was suspected to be related to the change (JECDB (2001)), it was classified in Category 2 (adrenal gland). Besides, there were increased liver weights in males at or above 50 mg/kg and females at 200 mg/kg, and centrilobular hypertrophy of hepatocytes was found histologically, but it was not adopted for classification because it is described that such centrilobular hypertrophy of hepatocytes is known to occur related to the induction of drug-metabolizing enzymes, and it was estimated to occur through a similar mechanism in this test (JECDB (2001)).
